目的:通过建立家犬膝骨关节炎(osteoarthritis,OA)模型,比较3种低浓度医用臭氧关节腔内注射对0A治疗的最适浓度。方法健康雄性犬40只,随机分为A组(空气组)、B组(10 ug/ml医用臭氧组)、C组(20 ug/ml医用臭氧组)、D组(30 ug/ml医用臭氧组),每组10只,左膝关节10只为N组(正常对照组)。采用Hulth制模,分别于造模前、造模成功时和处死前测定膝关节活动度,用硝酸还原酶法检测关节液中TNF-α含量。结果8周后A组膝关节活动度显著降低,B、C、D组治疗结束后较造模成功时增加,但C组较B、D组活动度明显增加( P<0.05);4组右侧膝关节均出现关节软骨病理损害, A组最严重,C组软骨病变最轻;关节液中TNF-α含量C组比A、B、D组明显降低,有统计学意义(P<0.05)。结论20μg/ml臭氧组能够减轻炎症反应,增强机体清除自由基的能力,抑制TNF-α细胞因子表达,从而减轻退行性关节炎软骨退变。

목적:통과건립가견슬골관절염(osteoarthritis,OA)모형,비교3충저농도의용취양관절강내주사대0A치료적최괄농도。방법건강웅성견40지,수궤분위A조(공기조)、B조(10 ug/ml의용취양조)、C조(20 ug/ml의용취양조)、D조(30 ug/ml의용취양조),매조10지,좌슬관절10지위N조(정상대조조)。채용Hulth제모,분별우조모전、조모성공시화처사전측정슬관절활동도,용초산환원매법검측관절액중TNF-α함량。결과8주후A조슬관절활동도현저강저,B、C、D조치료결속후교조모성공시증가,단C조교B、D조활동도명현증가( P<0.05);4조우측슬관절균출현관절연골병리손해, A조최엄중,C조연골병변최경;관절액중TNF-α함량C조비A、B、D조명현강저,유통계학의의(P<0.05)。결론20μg/ml취양조능구감경염증반응,증강궤체청제자유기적능력,억제TNF-α세포인자표체,종이감경퇴행성관절염연골퇴변。
Objective To compare the three low concentrationss of medical ozone for treating knee osteoarthritis ( OA) through intra-articular injection using the established knee OA model in dogs.Methods Forty healthy male dogs were randomly divided into Group A ( the air group) , Group B ( the 10 ug/ml medical ozone group) , Group C ( the 20 ug/ml medical ozone group) , and group D (the 30 ug/ml medical ozone group), with 10 dogs in each group.The left knee joints were selected as Group N (the normal control group) .Hulth method was used to create OA model.The range of knee motion was measured before modeling, successful modeling, and before execution of the dogs.Nitrale reduetase ( NR) method was used to detect the concentration of TNF-a in the joints.Results The range of knee motion in Group A decreased obviously after 8 weeks.The increased was more in Group B, C, and D after the therapy than that at the successful modeling.The range of knee motion in Group C increased more obviously than that in Group B and D (P<0.05).Articular cartilage pathological damage was found in the right knees of the dogs in 4 groups, and the most serious cartilage lesion was in Group A and the lest in group C.TNF-a concentration in Group C decrased more obviously than that in Group A, B, and D, which was of statistical difference ( P<0.05 ) .Conclusion The 20 ug/ml medical ozone can reduce inflammatory reaction, enhance the ability to eliminate free radicals of the body, and inhibit the expression of TNF-a.
