目的:本研究通过建立鼠肝脏缺血再灌注损伤模型,探索东莨菪碱应用于肝脏缺血再灌注损伤保护机制。方法(1)SD 大鼠60只均等随机分为 A、B、C 3组:A 组为假手术组,B组为缺血再灌注组,C 组为东莨菪碱组;(2)各组分别在4个时点(T1再灌注0 min,T230 min, T360 min,T4120 min)取血5 ml,测定血清中丙氨酸氨基转移酶(ALT)、天冬氨酸氨基转移酶(AST)、肿瘤坏死因子α(TNF-α)、白细胞介素10(IL-10)、黄嘌呤氧化酶(XOD)及丙二醛(MDA)含量;(3)取肝脏行病理检查。结果(1)B 组血清中 ALT、AST、MDA、XOD、TNF-α含量于4个时点均最高(P<0.05),且随再灌注时间延长而升高(P<0.05);(2)IL-10 C 组含量最高(P<0.05),随再灌注时间延长而升高(P<0.05);(3)肝组织病理结果:各时点中,B 组损伤最重,其次为 C组,A 组均正常。结论(1)鼠肝脏缺血再灌注损伤可通过损伤自由基、促进 TNF-α合成,减少IL-10释放而损伤肝细胞。(2)东莨菪碱具有保肝作用。

목적:본연구통과건립서간장결혈재관주손상모형,탐색동랑탕감응용우간장결혈재관주손상보호궤제。방법(1)SD 대서60지균등수궤분위 A、B、C 3조:A 조위가수술조,B조위결혈재관주조,C 조위동랑탕감조;(2)각조분별재4개시점(T1재관주0 min,T230 min, T360 min,T4120 min)취혈5 ml,측정혈청중병안산안기전이매(ALT)、천동안산안기전이매(AST)、종류배사인자α(TNF-α)、백세포개소10(IL-10)、황표령양화매(XOD)급병이철(MDA)함량;(3)취간장행병리검사。결과(1)B 조혈청중 ALT、AST、MDA、XOD、TNF-α함량우4개시점균최고(P<0.05),차수재관주시간연장이승고(P<0.05);(2)IL-10 C 조함량최고(P<0.05),수재관주시간연장이승고(P<0.05);(3)간조직병리결과:각시점중,B 조손상최중,기차위 C조,A 조균정상。결론(1)서간장결혈재관주손상가통과손상자유기、촉진 TNF-α합성,감소IL-10석방이손상간세포。(2)동랑탕감구유보간작용。
Objective To study the protective effect of scopolamine on hepatic ischemia reperfusion injury in rats. Methods Equal 60 SD rats were randomly divided into 3 groups named A, B and C. Group A was sham-operated group; Group B was the group of ischemia reperfusion; Group C was the group used scopolamine; In each group blood sampling was took for 5 ml on 4 points (respectively, in T1 for 0 min, T2 for 30 min, T3 for 60 min, T4 for 120 min), and were used to determine the contents of ALT, AST, TNF-α, IL-10, XOD, MDA3 in serum, perform pathological examination of the liver. Results The ALT, AST, MDA and the XOD, TNF-α levels of B Group serum were the highest in each points (P<0.05), and increased with prolonged reperfusion (P<0.05). IL-10 in group C was maximum (P<0.05), increased with prolonged reperfusion (P<0.05). Liver biopsy results: at each time point Group B was the most heavily damaged, followed by Group C, group A was normal. Conclusion Liver injury induced by ischemia-reperfusion injury via radicals and promoting the synthesis of TNF-α, IL-10 release can damage liver cells. Scopolamine has hepatoprotective effect.
