目的:观察四神丸对溃疡性结肠炎(UC)大鼠白细胞介素(IL)-1β、-4肿瘤坏死因子(TNF)-α的影响。方法 Wistar 大鼠40只分为四组,采用2、4、6-三硝基苯磺酸(TNBS)乙醇灌肠法造模,光镜观察结肠组织病理形态并评分,采用酶联免疫法( ELISA)检测大鼠血清 IL-1β、 TNF-α、IL-4的水平。结果模型组大鼠结肠组织黏膜层可见溃疡形成,且有明显的炎症、充血存在,证实造模成功。模型组大鼠血清 IL-1β、TNF-α含量均高于空白组(P<0.05),而 IL-4含量低于空白组(P<0.01);治疗后,四神丸组血清 IL-1β、TNF-α水平均较模型组降低(P<0.05、P<0.01),而 IL-4含量却显著升高(P<0.01)。结论四神丸能够通过下调 IL-1β、TNF-α的表达,升高 IL-4的表达,调节肠道异常免疫反应、抑制炎症反应,修复溃疡,进而治疗 UC。
目的:觀察四神汍對潰瘍性結腸炎(UC)大鼠白細胞介素(IL)-1β、-4腫瘤壞死因子(TNF)-α的影響。方法 Wistar 大鼠40隻分為四組,採用2、4、6-三硝基苯磺痠(TNBS)乙醇灌腸法造模,光鏡觀察結腸組織病理形態併評分,採用酶聯免疫法( ELISA)檢測大鼠血清 IL-1β、 TNF-α、IL-4的水平。結果模型組大鼠結腸組織黏膜層可見潰瘍形成,且有明顯的炎癥、充血存在,證實造模成功。模型組大鼠血清 IL-1β、TNF-α含量均高于空白組(P<0.05),而 IL-4含量低于空白組(P<0.01);治療後,四神汍組血清 IL-1β、TNF-α水平均較模型組降低(P<0.05、P<0.01),而 IL-4含量卻顯著升高(P<0.01)。結論四神汍能夠通過下調 IL-1β、TNF-α的錶達,升高 IL-4的錶達,調節腸道異常免疫反應、抑製炎癥反應,脩複潰瘍,進而治療 UC。
목적:관찰사신환대궤양성결장염(UC)대서백세포개소(IL)-1β、-4종류배사인자(TNF)-α적영향。방법 Wistar 대서40지분위사조,채용2、4、6-삼초기분광산(TNBS)을순관장법조모,광경관찰결장조직병리형태병평분,채용매련면역법( ELISA)검측대서혈청 IL-1β、 TNF-α、IL-4적수평。결과모형조대서결장조직점막층가견궤양형성,차유명현적염증、충혈존재,증실조모성공。모형조대서혈청 IL-1β、TNF-α함량균고우공백조(P<0.05),이 IL-4함량저우공백조(P<0.01);치료후,사신환조혈청 IL-1β、TNF-α수평균교모형조강저(P<0.05、P<0.01),이 IL-4함량각현저승고(P<0.01)。결론사신환능구통과하조 IL-1β、TNF-α적표체,승고 IL-4적표체,조절장도이상면역반응、억제염증반응,수복궤양,진이치료 UC。
Objective To observe the influence of the Si Shen Pill on interleukin -1β(IL-1β) ,TNF-αand interleukin-4(IL-4)in ulcerative colitis (UC) rats.Methods Experimental rats were divided into four groups .Model rats were made by means of the TNBS /ethanol enema.Colon organization pathological changes were observed by the light microscope .Serum IL-1β,TNF-αand IL-4 were detected by ELISA.Results Ulceration,inflammation,congestion existed in colon mucosa of model rats ,which meant the models were made successful -ly.IL-1βand TNF-αof model rats were higher and IL-4 was lower than those of rats in blank group (P<0.05,P<0.01).After treatment, compared with the model group,IL-1βand TNF-αwere reduced and IL-4 was increased in the Si Shen Pill group (P<0.05,P<0.01).Con-clusions Si Shen Pill can reduce the IL-1βand TNF-α,increase IL-4,regulate immune abnormalities in intestines ,inhibit inflammation,re-pair ulcers and cure UC.
