目的:探讨吸入麻醉药异氟烷对转 APP 基因小鼠学习记忆功能的影响及海藻糖的干预保护作用。方法60只12月龄转 APP 小鼠随机分为空白对照组(Control 组)、异氟烷组(Iso 组)、海藻糖组(Iso+Tre 组),每组20只。 Iso+Tre 组和 Iso 组分别于麻醉前30 min 腹腔注射海藻糖400μg /kg 或等量生理盐水,然后给予1.4%异氟烷吸入麻醉2 h,麻醉后24 h 应用 Morris 水迷宫测试小鼠学习记忆能力,麻醉后6、24 h 杀鼠取脑,采用 TUNEL 染色观察各组小鼠海马区神经元凋亡,应用 ELISA 法检测 Aβ含量,Western 印迹法检测 Bcl-2、Bax、Caspase-3的表达变化。结果与 Con-trol 组相比,Iso 组小鼠总体的平均逃避潜伏期明显延长,空间探索时间明显缩短( P <0.01),海马神经元凋亡指数、Aβ含量、Bax/Bcl-2比值及Caspase-3表达均明显增加(P<0.01);Iso+Tre 组与 Iso 组相比,逃避潜伏期缩短,空间探索时间延长(P<0.05),Aβ含量、Bax /Bcl-2比值及 Caspase-3表达均明显减少(P<0.01)。结论海藻糖能够改善异氟烷诱导转 APP 基因小鼠学习记忆能力的损害,减少海马神经元的凋亡及 Aβ的生成。

목적:탐토흡입마취약이불완대전 APP 기인소서학습기억공능적영향급해조당적간예보호작용。방법60지12월령전 APP 소서수궤분위공백대조조(Control 조)、이불완조(Iso 조)、해조당조(Iso+Tre 조),매조20지。 Iso+Tre 조화 Iso 조분별우마취전30 min 복강주사해조당400μg /kg 혹등량생리염수,연후급여1.4%이불완흡입마취2 h,마취후24 h 응용 Morris 수미궁측시소서학습기억능력,마취후6、24 h 살서취뇌,채용 TUNEL 염색관찰각조소서해마구신경원조망,응용 ELISA 법검측 Aβ함량,Western 인적법검측 Bcl-2、Bax、Caspase-3적표체변화。결과여 Con-trol 조상비,Iso 조소서총체적평균도피잠복기명현연장,공간탐색시간명현축단( P <0.01),해마신경원조망지수、Aβ함량、Bax/Bcl-2비치급Caspase-3표체균명현증가(P<0.01);Iso+Tre 조여 Iso 조상비,도피잠복기축단,공간탐색시간연장(P<0.05),Aβ함량、Bax /Bcl-2비치급 Caspase-3표체균명현감소(P<0.01)。결론해조당능구개선이불완유도전 APP 기인소서학습기억능력적손해,감소해마신경원적조망급 Aβ적생성。
Objective To observe the influence of inhalation anesthestic isoflurane on the learning -memory ability of APP transgenic mice and protective effects of trehalose .Methods 60 APP transgenic mice at the age of 12 months were divided into control , Iso and Iso+Tre groups.The abilities of learning and memory were investigated by Morris water maze .TUNEL assay was employed to observe the hipp -ocampal neuron apoptosis of the mice in each group .The amount of Aβwas checked by ELISA assay kit and the variations of Bcl -2, Bax, Caspase-3 were examined by Western blot.Results In comparison with those of control group , the mean escape latency of Iso group was prolonged, the spatial exploring time was reduced , apoptotic index, the amount of Aβ, the ratio of Bax/Bcl-2, and the expression of Caspase-3 were increased.Moreover, compared to those of Iso group, escape latency of Iso +Tre was shorter, longer spatial exploring time was longer, the amount of Aβ, ratio of Bax /Bcl-2, and Caspase-3 expression were lower.Conclusions Trehalose could improve the recover -y of the learning and memory of APP transgenic mice after isoflurane anesthesia , the hippocampal neuron apoptosis and the production of A βcan be inhibited by trehalose.
