目的:探讨香烟烟雾提取物(CSE)对Ⅱ型肺泡上皮细胞中 Derlin-1蛋白及其 mRNA 表达的影响。方法体外培养Ⅱ型肺泡上皮细胞(A549),加入10%浓度的 CES 作用于细胞0、3、6、12、24、48 h,采用免疫蛋白印迹(WB)检测 Derlin-1蛋白表达水平的变化,并运用逆转录聚合酶链反应(RT-PCR)检测 Derlin-1 mRNA 含量的变化。结果10%浓度的 CSE 作用下,Derlin-1蛋白的表达逐渐提高,并在6 h 达最高,随后逐渐下降, Derlin-1 mRNA 含量变化与 Derlin-1蛋白表达一致。结论香烟烟雾可影响Ⅱ型肺泡上皮细胞中 Derlin-1表达的变化,可能与 Derlin-1是内质网应激诱导凋亡的抑制剂有关。
目的:探討香煙煙霧提取物(CSE)對Ⅱ型肺泡上皮細胞中 Derlin-1蛋白及其 mRNA 錶達的影響。方法體外培養Ⅱ型肺泡上皮細胞(A549),加入10%濃度的 CES 作用于細胞0、3、6、12、24、48 h,採用免疫蛋白印跡(WB)檢測 Derlin-1蛋白錶達水平的變化,併運用逆轉錄聚閤酶鏈反應(RT-PCR)檢測 Derlin-1 mRNA 含量的變化。結果10%濃度的 CSE 作用下,Derlin-1蛋白的錶達逐漸提高,併在6 h 達最高,隨後逐漸下降, Derlin-1 mRNA 含量變化與 Derlin-1蛋白錶達一緻。結論香煙煙霧可影響Ⅱ型肺泡上皮細胞中 Derlin-1錶達的變化,可能與 Derlin-1是內質網應激誘導凋亡的抑製劑有關。
목적:탐토향연연무제취물(CSE)대Ⅱ형폐포상피세포중 Derlin-1단백급기 mRNA 표체적영향。방법체외배양Ⅱ형폐포상피세포(A549),가입10%농도적 CES 작용우세포0、3、6、12、24、48 h,채용면역단백인적(WB)검측 Derlin-1단백표체수평적변화,병운용역전록취합매련반응(RT-PCR)검측 Derlin-1 mRNA 함량적변화。결과10%농도적 CSE 작용하,Derlin-1단백적표체축점제고,병재6 h 체최고,수후축점하강, Derlin-1 mRNA 함량변화여 Derlin-1단백표체일치。결론향연연무가영향Ⅱ형폐포상피세포중 Derlin-1표체적변화,가능여 Derlin-1시내질망응격유도조망적억제제유관。
Objective To explore the effect of cigarette smoke extract (CSE) on expressional levels of Derlin-1 protein and mRNA in the cultured A549 cells those were rat alveolar type Ⅱ cells lines.Methods Western blot was used to detect the level of Derlin -1 protein expression exposured to 10% concentration of CSE at different time (0,3,6,12,24,48 h).RT-PCR was used to measure the change of Der -lin-1 mRNA.Results In 10% concentration of CSE, Derlin-1 protein expression gradually was increased and reached the highest at 6 h and then decreased gradually.RT-PCR results consistented with the result of Western blot .Conclusions Cigarette smoke can affect the changes of the expression of Derlin-1 in alveolar typeⅡ cells, which may be associated with that Derlin -1 is the apoptosis inhibitor of endoplasmic re -ticulum stress.
