目的:对白芷中有效成分欧前胡素和异欧前胡素进行筛选和鉴定,并考察其对大鼠血管活性的影响。方法利用细胞膜色谱法(CMC)和气质联用法(GC/MS)对白芷中的有效成分进行筛选和鉴定,筛选过程采用大鼠动脉细胞膜色谱柱(50 mm ×1.0mm),37℃条件下用pH7.4的磷酸钠缓冲液作为流动相,通过大鼠离体腹主动脉环实验对活性成分进行研究。结果欧前胡素和异欧前胡素在大鼠动脉细胞膜制成的细胞膜色谱系统中,能够产生与维拉帕米类似的药理作用。这2种成分能够显著抑制由重酒石酸去甲肾上腺素(NE)和氯化钙(CaCl2)诱导的血管收缩作用。结论在用白芷治疗血管疾病过程中,欧前胡素和异欧前胡素将作为2种主要发挥药理作用的有效成分。
目的:對白芷中有效成分歐前鬍素和異歐前鬍素進行篩選和鑒定,併攷察其對大鼠血管活性的影響。方法利用細胞膜色譜法(CMC)和氣質聯用法(GC/MS)對白芷中的有效成分進行篩選和鑒定,篩選過程採用大鼠動脈細胞膜色譜柱(50 mm ×1.0mm),37℃條件下用pH7.4的燐痠鈉緩遲液作為流動相,通過大鼠離體腹主動脈環實驗對活性成分進行研究。結果歐前鬍素和異歐前鬍素在大鼠動脈細胞膜製成的細胞膜色譜繫統中,能夠產生與維拉帕米類似的藥理作用。這2種成分能夠顯著抑製由重酒石痠去甲腎上腺素(NE)和氯化鈣(CaCl2)誘導的血管收縮作用。結論在用白芷治療血管疾病過程中,歐前鬍素和異歐前鬍素將作為2種主要髮揮藥理作用的有效成分。
목적:대백지중유효성분구전호소화이구전호소진행사선화감정,병고찰기대대서혈관활성적영향。방법이용세포막색보법(CMC)화기질련용법(GC/MS)대백지중적유효성분진행사선화감정,사선과정채용대서동맥세포막색보주(50 mm ×1.0mm),37℃조건하용pH7.4적린산납완충액작위류동상,통과대서리체복주동맥배실험대활성성분진행연구。결과구전호소화이구전호소재대서동맥세포막제성적세포막색보계통중,능구산생여유랍파미유사적약리작용。저2충성분능구현저억제유중주석산거갑신상선소(NE)화록화개(CaCl2)유도적혈관수축작용。결론재용백지치료혈관질병과정중,구전호소화이구전호소장작위2충주요발휘약리작용적유효성분。
Objective Effective components ,imperatorin and isoimperatorin ,from Radix A ngelicae dahuricae were screened and i‐dentified by using cell membrane chromatography (CMC) and gas chromatography/mass spectrometry (GC/MS) .Methods The components showed the effects of inhibiting vasoconstriction in vitro on rat abdominal aorta segments .The screening procedure was performed on a rat artery cell membrane column (50 mm × 1 .0 mm) with sodium phosphate buffer (pH 7 .4) as the mobile phase under temperature of 37 ℃ .Results The pharmacological results demonstrated that imperatorin and isoimperatorin can act on rat artery cell membrane as verapamil in CMC system .They can significantly inhibit the vasoconstrictions induced by norepi‐nephrine bitartrate (NE) and the calcium chloride (CaCl2 ) .Conclusion Imperatorin and isoimperatorin are two main effective components of Radix A ngelicae dahuricae as a medicine in the treatment for vessel disease .
