探讨当归多糖对麻黄素小鼠肝组织抗氧化酶活性和核转录因子‐κB(NF‐κB)、肿瘤坏死因子(TNF‐α)表达的影响及其可能作用机制。小鼠连续腹腔注射4?0 g?L -1麻黄素溶液,同时用当归多糖溶液(12?5,25?0 g?L -1)进行灌胃,分别在5,10,15 d时用分光光度法检测各组小鼠肝组织 T‐AOC、GSH‐PX活性的变化,免疫组化法检测小鼠肝组织中NF‐κB和TNF‐α表达的变化。结果显示实验对照组小鼠肝组织 T‐AOC、GSH‐PX活性明显降低(P<0?01),肝组织NF‐κB和TNF‐α表达量显著增强(P<0?05或 P<0?01),而当归多糖组 T‐AOC、GSH‐PX活性与实验对照组相比显著升高,NF‐κB和TNF‐α表达量明显降低(P<0?05或 P<0?01)。结果表明,麻黄素影响小鼠肝组织抗氧化酶活性和NF‐κB、TNF‐α的表达,当归多糖能够提高小鼠肝组织抗氧化酶活性,抑制NF‐κB和 TNF‐α的表达,对麻黄素致小鼠肝损伤有一定的保护作用。
探討噹歸多糖對痳黃素小鼠肝組織抗氧化酶活性和覈轉錄因子‐κB(NF‐κB)、腫瘤壞死因子(TNF‐α)錶達的影響及其可能作用機製。小鼠連續腹腔註射4?0 g?L -1痳黃素溶液,同時用噹歸多糖溶液(12?5,25?0 g?L -1)進行灌胃,分彆在5,10,15 d時用分光光度法檢測各組小鼠肝組織 T‐AOC、GSH‐PX活性的變化,免疫組化法檢測小鼠肝組織中NF‐κB和TNF‐α錶達的變化。結果顯示實驗對照組小鼠肝組織 T‐AOC、GSH‐PX活性明顯降低(P<0?01),肝組織NF‐κB和TNF‐α錶達量顯著增彊(P<0?05或 P<0?01),而噹歸多糖組 T‐AOC、GSH‐PX活性與實驗對照組相比顯著升高,NF‐κB和TNF‐α錶達量明顯降低(P<0?05或 P<0?01)。結果錶明,痳黃素影響小鼠肝組織抗氧化酶活性和NF‐κB、TNF‐α的錶達,噹歸多糖能夠提高小鼠肝組織抗氧化酶活性,抑製NF‐κB和 TNF‐α的錶達,對痳黃素緻小鼠肝損傷有一定的保護作用。
탐토당귀다당대마황소소서간조직항양화매활성화핵전록인자‐κB(NF‐κB)、종류배사인자(TNF‐α)표체적영향급기가능작용궤제。소서련속복강주사4?0 g?L -1마황소용액,동시용당귀다당용액(12?5,25?0 g?L -1)진행관위,분별재5,10,15 d시용분광광도법검측각조소서간조직 T‐AOC、GSH‐PX활성적변화,면역조화법검측소서간조직중NF‐κB화TNF‐α표체적변화。결과현시실험대조조소서간조직 T‐AOC、GSH‐PX활성명현강저(P<0?01),간조직NF‐κB화TNF‐α표체량현저증강(P<0?05혹 P<0?01),이당귀다당조 T‐AOC、GSH‐PX활성여실험대조조상비현저승고,NF‐κB화TNF‐α표체량명현강저(P<0?05혹 P<0?01)。결과표명,마황소영향소서간조직항양화매활성화NF‐κB、TNF‐α적표체,당귀다당능구제고소서간조직항양화매활성,억제NF‐κB화 TNF‐α적표체,대마황소치소서간손상유일정적보호작용。
The effect of Angelica sinensis polysaccharide on activities of antioxidant enzyme and expressions of NF‐κB、 TNF‐αin liver tissue of the mice injured by ephedrine are investigated , and its possible mechanism is also explored . The mice are intraperitoneal injected 4?0 g?L -1 ephedrine , and irrigated to stomach with A ngelica sinensis polysaccharide solution (12?5 ,25?0 g?L -1 ) after injected ephedrine 1 h later . The samples at 5 ,10 and 15 d respectively ,activities of hepatic histological T‐AOC、GSH‐PX are detected by spectrophotometer . The expressions of NF‐κB and TNF‐αare measured by immunohistochemistry in liver of mice at different developmental stages . Results show the activities of hepatic histological T‐AOC、GSH‐PX of experiment control group significantly reduce (P< 0?01) , the positive expressions of NF‐κB and TNF‐αof live obviously increase(P<0?05 or P<0?01);Compared with experiment control group , the activities of T‐AOC、 GSH‐PX increase significantly and the positive expressions of NF‐κB and TNF‐αof liver significantly reduce of Angelica sinensis polysaccharide group(P <0?05 or P< 0?01 ) . Indicate A ngelica sinensis polysaccharide can improve liver tissue antioxidant capacity and inhibiting the expressions of NF‐κB and TNF‐α, which may be one of the important mechanisms for the treatment of liver injury by ephedrine .
