Qiyuan Yishen granule%adriamycin nephrosis( AN)%T-lymphocyte subsets%CD28%rats
目的:观察芪苑益肾颗粒对阿霉素肾病大鼠外周血T细胞亚群及共刺激分子CD28表达的影响。方法采用单次尾静脉注射阿霉素14 d制造肾病大鼠模型,空白组注射等量生理盐水。造模成功后,空白对照组和模型对照组分别给予蒸馏水,而阳性对照(醋酸泼尼松片5 mg/kg)、芪苑益肾颗粒低(1.5 g/kg)、中(3.0 g/kg)、高剂量(6.0 g/kg)分别给药,连续灌胃21 d后,采用流式细胞术检测阿霉素肾病大鼠外周血T细胞亚群CD3+、CD4+、CD8+、CD4/CD8及共刺激分子CD28的表达。结果外周血T细胞亚群CD3+、CD4+芪苑益肾颗粒低、中、高剂量组显著高于阿霉素肾病大鼠模型组(P<0.05);共刺激分子CD28的表达低、中剂量显著高于模型组(P<0.01);与模型组比较,高剂量组CD4/CD8显著升高(P<0.01),而CD8+显著降低(P<0.05)。结论芪苑益肾颗粒可能对阿霉素肾病大鼠外周血T细胞亚群及共刺激分子CD28表达有免疫调节作用。
目的:觀察芪苑益腎顆粒對阿黴素腎病大鼠外週血T細胞亞群及共刺激分子CD28錶達的影響。方法採用單次尾靜脈註射阿黴素14 d製造腎病大鼠模型,空白組註射等量生理鹽水。造模成功後,空白對照組和模型對照組分彆給予蒸餾水,而暘性對照(醋痠潑尼鬆片5 mg/kg)、芪苑益腎顆粒低(1.5 g/kg)、中(3.0 g/kg)、高劑量(6.0 g/kg)分彆給藥,連續灌胃21 d後,採用流式細胞術檢測阿黴素腎病大鼠外週血T細胞亞群CD3+、CD4+、CD8+、CD4/CD8及共刺激分子CD28的錶達。結果外週血T細胞亞群CD3+、CD4+芪苑益腎顆粒低、中、高劑量組顯著高于阿黴素腎病大鼠模型組(P<0.05);共刺激分子CD28的錶達低、中劑量顯著高于模型組(P<0.01);與模型組比較,高劑量組CD4/CD8顯著升高(P<0.01),而CD8+顯著降低(P<0.05)。結論芪苑益腎顆粒可能對阿黴素腎病大鼠外週血T細胞亞群及共刺激分子CD28錶達有免疫調節作用。
목적:관찰기원익신과립대아매소신병대서외주혈T세포아군급공자격분자CD28표체적영향。방법채용단차미정맥주사아매소14 d제조신병대서모형,공백조주사등량생리염수。조모성공후,공백대조조화모형대조조분별급여증류수,이양성대조(작산발니송편5 mg/kg)、기원익신과립저(1.5 g/kg)、중(3.0 g/kg)、고제량(6.0 g/kg)분별급약,련속관위21 d후,채용류식세포술검측아매소신병대서외주혈T세포아군CD3+、CD4+、CD8+、CD4/CD8급공자격분자CD28적표체。결과외주혈T세포아군CD3+、CD4+기원익신과립저、중、고제량조현저고우아매소신병대서모형조(P<0.05);공자격분자CD28적표체저、중제량현저고우모형조(P<0.01);여모형조비교,고제량조CD4/CD8현저승고(P<0.01),이CD8+현저강저(P<0.05)。결론기원익신과립가능대아매소신병대서외주혈T세포아군급공자격분자CD28표체유면역조절작용。
Objective To explore the effect of Qiyuan Yishen granule on the T-lymphocyte subsets in peripheral blood and the ex-pression of costimulatory molecule CD28 of adriamycin nephrosis( AN) rats. Methods The rat nephropathy model was established by the single intravenous injection of adriamycin via tail for 14 d. The control rats were only injected with the same amount of saline. After the success of modeling,the rats were given the same amount of distilled water by gavage in normal control group and model group,while the rats were continuously administered with drug by gastric tube for 21 d in positive control group(5 mg/kg),Qiyuan Yishen granule low dose group(1. 5 g/kg),middle dose group(3. 0 g/kg),high dose group(6. 0 g/kg). The levels of CD3+,CD4+,CD8+, CD4/CD8 and the expression of costimulatory molecule CD28 in peripheral blood of adriamycin nephrosis rats were detected using flow cytom-etry. Results The levels of CD4+,CD3+ in peripheral blood were significantly higher in three Qiyuan Yishen granule groups than in model group(P<0. 05). The expression of costimulatory molecule CD28 in low,medium dose of Qiyuan Yishen granule groups were significantly higher than in model group(P<0. 01). The level of CD4/CD8 in high dose group was significantly higher than in model group(P<0. 01), while the level of CD8+was significantly lower than in model group(P<0. 05). Conclusion Qiyuan Yishen granule may have a certain immune regulation effect on the T-lymphocyte subsets in peripheral blood and the expression of costimulatory molecule CD28 of adria-mycin nephrosis( AN) rats.
