目的:观察脑源性神经营养因子(BDNF)预处理给药对癫痫家兔的干预效应及其与神经肽Y(NPY)表达的关系。方法雄性家兔侧脑室注射红藻氨酸(KA)建立癫痫动物模型,将72只家兔随机分为假手术组、BDNF对照组、癫痫模型组、BDNF预处理组,各组再分为造模成功后6 h、12 h及24 h处死3个亚组,每个亚组6只家兔。采用免疫组织化学技术测定海马组织中NPY蛋白的表达。结果 BDNF预处理组家兔癫痫发作级别及神经元损害较癫痫模型组降低;癫痫模型组3个亚组的大鼠海马组织中NPY蛋白表达较假手术组相对应的各亚组明显增多(P<0.05);BDNF预处理组中3个亚组家兔海马组织中NPY蛋白的表达较癫痫模型组相对应的各亚组明显增多(P<0.05);BDNF对照组与癫痫模型组相比相关指标均无统计学意义;同组内6h、12h、24h 3个时间点NPY蛋白表达呈上升趋势。结论BDNF预处理给药可上调NPY蛋白表达,减轻神经元的损害,从而在一定程度上发挥抗癫痫发作的药理作用。

Objective To investigate the intervention effects of brain-derived neutrophic factor (BDNF) pretreatment on epileptic rabbits and its relationship with the expression of neuropeptide Y (NPY).Methods An animal model of complex partial epilepsy was established in male rabbits by intracerebroventricular injection of Kainic acid. 72 rabbits rats were randomly divided into sham operation group, BDNF control group, epilepsy model group, and BDNF pretreatment group. Each group was further divided into three subgroups(6 hours, 12 h and 24 h after seisures). Morphologic changes of the hippocampal neurons were observed by HE staining, while the expressions of NPY in hippocampal tissue were measured by immunohistochemical techniques.Results Seizure levels and neuronal damage of rats in BDNF pretreatment group were lower than those in epilepsy model group. In hippocampal tisse, NPY expression of three subgroups in epilepsy model group increased significantly than those in sham operation group (P<0.05); NPY expressions of three subgroups of BDNF pretreatment group increased significantly than those in epilepsy model group (P<0.05). No statistically different of NPY expression was found between BDNF control group and epilepsy model group (P>0.05). Upturns of NPY expression was showed from 6 h to 24 h time point after seisures in the same group.Conclusion BDNF pretreatment have some antiepileptic effects which are concerned with lessen of neuronic damage, the mitigation of seizure degree, and the up-regulation of NPY expression.
