目的探讨高糖及雷公藤内酯醇(TP)对人肾小球系膜细胞单核细胞趋化蛋白-1(MCP-1)表达的影响。方法将培养的人肾小球系膜细胞分为6组:正常对照组(5 mmol/L 葡萄糖)、高糖组(30 mmol/L 葡萄糖)、甘露醇组(5 mmol/L 葡萄糖+25 mmol/L甘露醇)、TP甲组(30 mmol/L 葡萄糖+5μg/L TP)、TP 乙组(30 mmol/L 葡萄糖+10μg/L TP)、TP丙组(30mmol/L 葡萄糖+15μg/L TP),分别在24 h、48 h、72 h采用 RT-PCR法检测TP对高糖条件下系膜细胞内 MCP-1 mRNA 及蛋白表达的影响。结果高糖组人肾小球系膜细胞 MCP-1 mRNA及蛋白较正常对照组增加,且差异有显著性(P<0.01);TP各组比高糖组有明显下降(P<0.01);不同浓度的TP对高糖诱导的系膜细胞 MCP-1的表达抑制程度不同(P <0.05)。结论 TP可抑制高糖诱导的人肾小球系膜细胞 MCP-1的表达,且呈时间、剂量依赖关系,其可能对延缓糖尿病肾病中肾小球硬化有一定作用。
目的探討高糖及雷公籐內酯醇(TP)對人腎小毬繫膜細胞單覈細胞趨化蛋白-1(MCP-1)錶達的影響。方法將培養的人腎小毬繫膜細胞分為6組:正常對照組(5 mmol/L 葡萄糖)、高糖組(30 mmol/L 葡萄糖)、甘露醇組(5 mmol/L 葡萄糖+25 mmol/L甘露醇)、TP甲組(30 mmol/L 葡萄糖+5μg/L TP)、TP 乙組(30 mmol/L 葡萄糖+10μg/L TP)、TP丙組(30mmol/L 葡萄糖+15μg/L TP),分彆在24 h、48 h、72 h採用 RT-PCR法檢測TP對高糖條件下繫膜細胞內 MCP-1 mRNA 及蛋白錶達的影響。結果高糖組人腎小毬繫膜細胞 MCP-1 mRNA及蛋白較正常對照組增加,且差異有顯著性(P<0.01);TP各組比高糖組有明顯下降(P<0.01);不同濃度的TP對高糖誘導的繫膜細胞 MCP-1的錶達抑製程度不同(P <0.05)。結論 TP可抑製高糖誘導的人腎小毬繫膜細胞 MCP-1的錶達,且呈時間、劑量依賴關繫,其可能對延緩糖尿病腎病中腎小毬硬化有一定作用。
목적탐토고당급뢰공등내지순(TP)대인신소구계막세포단핵세포추화단백-1(MCP-1)표체적영향。방법장배양적인신소구계막세포분위6조:정상대조조(5 mmol/L 포도당)、고당조(30 mmol/L 포도당)、감로순조(5 mmol/L 포도당+25 mmol/L감로순)、TP갑조(30 mmol/L 포도당+5μg/L TP)、TP 을조(30 mmol/L 포도당+10μg/L TP)、TP병조(30mmol/L 포도당+15μg/L TP),분별재24 h、48 h、72 h채용 RT-PCR법검측TP대고당조건하계막세포내 MCP-1 mRNA 급단백표체적영향。결과고당조인신소구계막세포 MCP-1 mRNA급단백교정상대조조증가,차차이유현저성(P<0.01);TP각조비고당조유명현하강(P<0.01);불동농도적TP대고당유도적계막세포 MCP-1적표체억제정도불동(P <0.05)。결론 TP가억제고당유도적인신소구계막세포 MCP-1적표체,차정시간、제량의뢰관계,기가능대연완당뇨병신병중신소구경화유일정작용。
Obj ective]To explore the effect of high glucose and triptolide(TP)on the expression of mono-cyte chemoattractant protein-1(MCP-1)in human mesangial cells(HMCs).[Methods]The cultured human HMCs were divided into normal control group(5mmol/l glucose),high glucose group(30mmol/l glucose), mannitol group(5mmol/l glucose with 25mmol/l mannitol),TP A groups(30mmol/l glucose with 5ug/L TP), TP B group(30mmol/L glucose with 10ug/L TP)and TP C group(30mmol/L glucose with 15ug/L TP).The effect of TP on the mRNA and protein expression of MCP-1 in HMCs induced by high glucose at 24h,48h and 72h were measured by RT-PCR.[Results]Compared with normal control group,the mRNA and protein ex-pression of MCP-1 in HMCS of high glucose group were increased,and there was significant difference(P<0.01),but those in TP groups were obviously decreased(P<0.01).The effect of different concentrations of TP in inhibiting the expression of MCP-1 of HMCs induced by high glucose was varied(P<0.05).[Conclu-sion]TP can inhibit the expression of MCP-1 in HMCs induced by high glucose in time and dose-dependent manners,and may be helpful in delaying the development and progression of glomerular sclerosis of diabetic nephropathy.
