目的探讨沉默 Caveolin-1(Cav-1)基因对人膀胱癌细胞株5637增殖和凋亡的影响。方法通过siRNA转染人膀胱癌细胞株5637(沉默组),以转染 FAM-siRNA为阴性对照组,未转染细胞为正常对照组。Re-al-time PCR法检测Cav-1基因沉默的效率,CCK-8法检测各组细胞的增殖情况,流式细胞术检测各种细胞的凋亡情况。结果 siRNA转染可显著降低人膀胱癌细胞株5637中 Cav-1 mRNA 的表达水平。沉默组细胞的增强能力明显减低,而细胞凋亡率明显升高(均P<0.05)。结论沉默Cav-1基因可抑制人膀胱癌细胞株5637的细胞增殖并促进细胞凋亡。

목적탐토침묵 Caveolin-1(Cav-1)기인대인방광암세포주5637증식화조망적영향。방법통과siRNA전염인방광암세포주5637(침묵조),이전염 FAM-siRNA위음성대조조,미전염세포위정상대조조。Re-al-time PCR법검측Cav-1기인침묵적효솔,CCK-8법검측각조세포적증식정황,류식세포술검측각충세포적조망정황。결과 siRNA전염가현저강저인방광암세포주5637중 Cav-1 mRNA 적표체수평。침묵조세포적증강능력명현감저,이세포조망솔명현승고(균P<0.05)。결론침묵Cav-1기인가억제인방광암세포주5637적세포증식병촉진세포조망。
Objective]To explore the effect of the silencing of caveolin-1(Cav-1)gene on the proliferation and apoptosis of human bladder cancer cell line 5637 in vitro.[Methods]Cav-1 gene was transfected into human blad-der cancer cell line 5637 by siRNA(silence group).Negative control group was transfected with FAM-siRNA, while normal control group was not transfected with FAM-siRNA.Real-time PCR was used to detect the efficacy of the silencing of CAV-1 gene.CCK-8 method was used to detect the cell proliferation in each group.Cell apopto-sis was measured by flow cytometry.[Results]SiRNA transfection could significantly reduce the mRNA expres-sion of Cav-1 in human bladder cancer cell line 5637.In the silence group,cell proliferation was obviously de-creased and cell apoptosis was obviously increased(all P<0.05).[Conclusion]The silencing of Cav-1 gene can in-hibit the proliferation and promote the apoptosis of human bladder cancer cells.
