目的:建立测定米粉中甲醛次硫酸氢钠(吊白块)含量的高效液相色谱法。方法:依据在酸性环境下中,样品中的甲醛次硫酸氢钠分解释放出的甲醛溶解到水中,提取后的甲醛与2,4-二硝基苯肼发生加成反应,生成黄色的2,4-二硝基苯腙,用正己烷萃取后,经高效液相色谱仪分离测定。结果:经优化试验确定样品振荡时间为20min,最佳衍生时间为30min,正己烷萃取时间为20min,利用高效液相色谱分离测定,试验条件确定为乙腈:水(60:40),流速为0.3mL/min,分离效果最好,利用标准甲醛衍生物保留时间对照定性,标准曲线定量测定次硫酸氢钠甲醛含量,该方法检出限为0.2mg/kg,回收率为94.6%~96.2%。结论:所建方法可用于米粉中的甲醛次硫酸氢钠的高效检测。

Objective:To establish a HPLC method for the content determination of sodium bisulfxylate formaldehyde in rice flour. Method:In acid solution, formaldehyde released from the decomposition of residual sodium bisulfxylate formaldehyde in the sample was extracted by water, then extracted formaldehyde and 2,4-dinitrophenylhydrazine (DNPH) took the addition reaction for genertating yellow 2,4-dinitrobenzene,which was separated by HPLC after extraction with n-hexane. Result:The best time of exact was 20 min, the act time was 30min ,the best time of extraction with n-hexane was 20 min. The best detecting condition was acetonitrile:water(60:40),the flow rate was 0.3mL/min,use the retention time of standard formaldehyde derivative as the control to calculate the content of sodium bisulfxylate formaldehyde. The detection limit was 0.2mg/kg in sample, and recovery was 94.6%~96.2%.Conclusion The establish method can be used in the test of sodium bisulfxylate formaldehyde in rice flour.
