目的:探讨赤芍水提物对肺癌细胞的抑制效应。方法选取人肺腺癌细胞A549、H1299以及小鼠Lewis肺腺癌细胞(LLC)为靶细胞,接种于6孔板或96孔板,接种密度为5×104/孔和3×103/孔,加入0、0.1、0.2、0.3、0.4、0.5、0.6、0.7、0.8、0.9、1.0 g/L不同浓度的赤芍水提物培养1~3 d,以0 g/L浓度的赤芍水提物处理组作为对照组,分别采用镜下观察、细胞计数仪以及细胞计数试剂盒(CCK-8)、流式细胞学检测细胞生长状态。结果镜下直接观察,与对照组(0 g/L浓度)比较,不同浓度的赤芍水提物处理后肺癌细胞数量明显较少,0.5 g/L浓度组细胞生长状态不佳,细胞增殖受到明显抑制,抑制率为43%(P<0.05)。流式细胞学检测显示,经赤芍水提物处理后,A549细胞周期阻滞于G1期。动物实验表明,赤芍可以抑制LLC皮下移植瘤的生长。结论赤芍中药可以抑制肺癌细胞的生长。

목적:탐토적작수제물대폐암세포적억제효응。방법선취인폐선암세포A549、H1299이급소서Lewis폐선암세포(LLC)위파세포,접충우6공판혹96공판,접충밀도위5×104/공화3×103/공,가입0、0.1、0.2、0.3、0.4、0.5、0.6、0.7、0.8、0.9、1.0 g/L불동농도적적작수제물배양1~3 d,이0 g/L농도적적작수제물처리조작위대조조,분별채용경하관찰、세포계수의이급세포계수시제합(CCK-8)、류식세포학검측세포생장상태。결과경하직접관찰,여대조조(0 g/L농도)비교,불동농도적적작수제물처리후폐암세포수량명현교소,0.5 g/L농도조세포생장상태불가,세포증식수도명현억제,억제솔위43%(P<0.05)。류식세포학검측현시,경적작수제물처리후,A549세포주기조체우G1기。동물실험표명,적작가이억제LLC피하이식류적생장。결론적작중약가이억제폐암세포적생장。
Objective To study the inhibition effect of aqueous extract from Pae onia v e itc hii on lung cancer cell growth. Methods A549, H1299, LLC cells were separately seeded in 6-well plates or 96-well plates. Different concen-tration (0, 0.1, 0.2, 0.3, 0.4, 0.5, 0.6, 0.7, 0.8, 0.9, 1.0 g/L) aqueous extract from Paeonia veitchii were added into cul-ture system, with 0 g/L concentration group as control group. At different time point, the cell morphology, proliferation and apoptosis were detected by microscopy, CCK-8 kit, flow cytometry. Results Compared to control group (0 g/L), the cell number of Pae onia v e itc hii treated group obviously decreased and the growth inhibitory effect was about 43% (P<0.05). The cell morphology was lost and cells cycle was inhibited in G1 phase. Animal experiments showed that the growth of LLC tumors could be inhibited by Paeonia veitchii. Conclusion Paeonia veitchii aqueous extract has a certain inhibitory effect on lung cancer cell growth.
