目的:研究肿瘤相关成纤维细胞( cancer associated fibroblasts, CAFs)对口腔鳞癌细胞生物学行为的影响,初步探讨CAFs在口腔鳞癌发生发展中的作用。方法采用原代培养的方法,培养口腔鳞癌相关成纤维细胞,光学显微镜观察其形态,免疫组化和免疫印迹实验进行鉴定,CCK-8法检测细胞的增殖能力。 transwell迁移实验检测CAFs诱导口腔鳞癌细胞迁移的能力;萤火虫荧光素酶报告系统检测CAFs对肿瘤细胞增殖能力的影响;平板克隆形成实验评价 CAFs对肿瘤细胞克隆形成能力的影响。结果成功分离培养出口腔鳞癌来源的 CAFs。CAFs能促进口腔鳞细胞的迁移、增殖和克隆形成等生物学行为。结论 CAFs能显著影响口腔鳞癌的增殖和迁移,表明其在口腔鳞癌的发生发展中发挥着一定的作用。

Objective To investigate the effect of cancer associated fibroblasts ( CAFs ) on the growth and migration of oral squamous cell carcinoma cells, and the possible role of CAFs in the tumorigenesis of oral squamous cell carcinoma. Methods CAFs were obtained from oral cancer tissues by primary culture. Its biological characteristics were identified by microscope observation, immunohistochemistry, western blotting and CCK-8 Cell Counting Kit. Transwell migration assay, luciferase reporter assay and plate clone formation assay were implemented to evaluate the influence of CAFs on oral squamous cell carcinoma cells. Results CAFs were isolated successfully from oral cancer tissues. Co-culture with CAFs promoted the growth, migration and colony forming ability of oral squamous cell carcinoma cells. Conclusion CAFs can obviously influence the growth and migration of oral squamous cell carcinoma cells,suggesting that CAFs may play a role in the development of oral squamous cell carcinoma.
