目的:探讨CBX7基因与膀胱癌转移的相关性及其过表达对膀胱癌细胞侵袭能力的影响。方法:应用荧光定量PCR检测膀胱癌组织中CBX7 mRNA的表达。构建CBX7真核表达载体pcDNA-CBX7,转染膀胱癌T24细胞,检测转染后CBX7蛋白的表达以及T24细胞的侵袭能力。结果:CBX7基因在膀胱癌组织中明显下调,而且侵袭性膀胱癌组织中CBX7的表达显著低于浅表性膀胱癌;pcDNA-CBX7转染后能够显著升高T24细胞CBX7的表达量;CBX7过表达的T24细胞穿透滤膜的数量明显减少。结论:CBX7基因与膀胱癌的转移相关,其过表达能够降低T24细胞的侵袭能力。

목적:탐토CBX7기인여방광암전이적상관성급기과표체대방광암세포침습능력적영향。방법:응용형광정량PCR검측방광암조직중CBX7 mRNA적표체。구건CBX7진핵표체재체pcDNA-CBX7,전염방광암T24세포,검측전염후CBX7단백적표체이급T24세포적침습능력。결과:CBX7기인재방광암조직중명현하조,이차침습성방광암조직중CBX7적표체현저저우천표성방광암;pcDNA-CBX7전염후능구현저승고T24세포CBX7적표체량;CBX7과표체적T24세포천투려막적수량명현감소。결론:CBX7기인여방광암적전이상관,기과표체능구강저T24세포적침습능력。
Objective:To research the relationship between CBX7 gene and the metastasis of bladder cancer and the influence of CBX7 over-expression to invasiveness of bladder cancer cells. Methods:Fluorescent quantitative PCR was used to detect the CBX7 mRNA in bladder cancer tissues. The expression vector pcDNA-CBX7 was con-structed and transfected to T24 cells. After transfection,invasiveness of T24 cells was detected. Results:The expres-sion level of CBX7 gene′s mRNA in bladder cancer tissue was significantly lower than that of para-cancerous tissue, and the expression level of CBX7 gene′s mRNA in invasive bladder cancer tissue was significantly lower than that of superficial bladder cancer. The pcDNA-CBX7 can significantly increase the expression of the CBX7 gene by Western blot. The over-expression of CBX7 gene can significantly inhibit the invasion ability of T24 cells. Conclusion:There is a relationship between CBX7 gene with metastasis of bladder cancer,and the over-expression of CBX7 gene can inhibit the invasiveness of T24 cells.
