为解决鲜食甜玉米多糖测定过程中玉米黄色素影响指示剂显色及水浴法提取多糖的提取率低等问题,建立了超声辅助水浴法提取多糖技术和苯酚-硫酸法检测技术,以葡萄糖为对照,测定速冻甜玉米粒中多糖含量。结果表明,苯酚-硫酸法测鲜食甜玉米多糖含量的精密度试验RSD值为1.296%,稳定性试验RSD值为1.675%,平均加样回收率为98.570%,RSD值为1.250%,测得鲜食甜玉米粒多糖含量为12.70%。此方法简便、准确率高、重现性较好。

To avoid the effect of maize yellow pigment on indicator in determining the polysaccharide of fresh sweet corn and resolve the problem of low extraction rate by water bath extraction method,the ultra-sonic-assisted extraction technology and the phenol-sulfuric acid detection technology were established in the study. The polysaccharide content of fresh sweet corn was then detected by using glucose as the stan-dard sample. The results showed that the RSD value in accuracy experiment and stability experiment was 1. 296% and 1. 675%,respectively. The average recovery rate was 98. 570%,with a RSD of 1. 250%. The content of polysaccharide in grains of fresh sweet corn was 12. 70%. This method was simple,highly accurate and well repeatable.
