目的 制备雷帕霉素(RAPA)脂质体滴眼液并研究RAPA对碱烧伤后大鼠角膜新生血管(corneal neovascularization,CNV)的抑制作用.方法 利用薄膜分散法制备RAPA脂质体滴眼液.采用成组设计法,将102只健康Wistar大鼠分组,碱烧伤后每日裂隙灯显微镜观察眼局部情况,并详细记录碱烧伤后大鼠角膜反应情况与新生血管生长情况,分别于碱烧伤后1d、4d、7d、14d随机抽取各组大鼠取材,行HE染色分析.实验结果采用方差分析及组间q检验进行分析.结果 (1)制得的RAPA脂质体,为完整的类球形,平均粒径为145.2 nm,包封率为90.02%,RAPA脂质体滴眼液中RAPA含量高,粒径合适,可作为滴眼液用于动物实验.(2) RAPA脂质体滴眼液组CNV生长迟缓、稀疏,退化较早;CNV面积较小,差异有统计学意义(P<0.05).结论 脂质体是RAPA良好的药物载体.RAPA能显著抑制CNV的生长,有望成为防治CNV的一种有效方法.
目的 製備雷帕黴素(RAPA)脂質體滴眼液併研究RAPA對堿燒傷後大鼠角膜新生血管(corneal neovascularization,CNV)的抑製作用.方法 利用薄膜分散法製備RAPA脂質體滴眼液.採用成組設計法,將102隻健康Wistar大鼠分組,堿燒傷後每日裂隙燈顯微鏡觀察眼跼部情況,併詳細記錄堿燒傷後大鼠角膜反應情況與新生血管生長情況,分彆于堿燒傷後1d、4d、7d、14d隨機抽取各組大鼠取材,行HE染色分析.實驗結果採用方差分析及組間q檢驗進行分析.結果 (1)製得的RAPA脂質體,為完整的類毬形,平均粒徑為145.2 nm,包封率為90.02%,RAPA脂質體滴眼液中RAPA含量高,粒徑閤適,可作為滴眼液用于動物實驗.(2) RAPA脂質體滴眼液組CNV生長遲緩、稀疏,退化較早;CNV麵積較小,差異有統計學意義(P<0.05).結論 脂質體是RAPA良好的藥物載體.RAPA能顯著抑製CNV的生長,有望成為防治CNV的一種有效方法.

Objective To prepare rapamycin liposomes gutta,and to evaluate their effect of inhibiting corneal neovascularization of rats induced by alkali cauterization.Methods Rapamycin liposomes were prepared by thin film hydration.Of 102 Wistar rats were divided into four groups at random.All right eyes of 96 rats were induced by alkali cauterization.After alkali burn,cauterized rats were observed by slitlamp biomicroscope every day.The analysis of variance and q test groups for analysis were adopted to analyze the results.Results The body of RAPA liposome was intact kinds of spheres,the average diameter was 145.2nm,and the envelopment rate was 90.02%.RAPA liposome could be used for animal's experiment as drip eyes liquid.The CNV of the rapamycin liposome treated group slowly and smaller than other group (P <0.05).Conclusions Liposome body is an excellent medicine carrier for the RAPA.RAPA can obviously suppress the growth of CNV.
