目的 观察振荡电场刺激(OFS)对脊髓少突胶质前体细胞(OPCs)活化为成熟少突胶质细胞及髓鞘再形成的影响.方法 48只SD大鼠采用Allen's打击法建立脊髓损伤模型,随机分为实验组和对照组.实验组植入刺激器给予振荡电场刺激,对照组只植入振荡电场刺激器但不施加刺激.在实验开始后的4d、7d、10d、14d取两组大鼠脊髓进行免疫荧光染色观察成熟少突胶质细胞的表达;4周和6周取大鼠脊髓行固蓝染色观察髓鞘形成情况,同时检测各组大鼠BBB功能学评分.结果 成熟少突胶质细胞的表达在4 ~14d时呈现递增的趋势,但实验组成熟少突胶质细胞在表达数量和体积上明显优于对照组,同时细胞排列更加有序;在6周时,实验组脊髓白质内的平均髓鞘面积相对比和BBB评分与对照组比较差异有统计学意义(P<0.05).而4周时,BBB评分两组之间差异有统计学意义,平均髓鞘面积相对比之间差异无统计学意义(P>0.05).结论 振荡电场刺激可以促进少突胶质前体细胞活化为成熟少突胶质细胞,促进髓鞘的再形成从而改善脊髓损伤后的神经功能恢复.并且,运动功能的改善较髓鞘的形成可以更早和明显.

Objective To evaluate the effects of oscillating field stimulation on the activation of oligodendrocyte precursor cells and remyelination after spinal cord injury.Methods Forty-eight SD rats with spinal cord injury induced by Allen' s method were randomly divided into experimental group and control group.The experimental group was implanted oscillating electrical field as intervention,while the control group only reveived oscillating electric field stimulator but not applied stimulus.At 4,7,10,14 d,4 w and 6 w after experiment,immunofluorescence staining,hematoxylin and eosin (HE) dyeing histology,BBB scores were used to analyze the expression of mature oligodendrocytes,remyelination and nerve function of SD rats.The statistical analysis about BBB scores and remyelination was performed between 2 groups.Results Mature oligodendrocytes expressed an increasing trend from 4 d to 14 d,while mature oligodendrocytes in the number and volume in the experimental group expressed better than that in the control group,and the cells are arranged in a more orderly.The average area of myelin and BBB (Basso-Beattie-Bresnahan) scores both presented a significant difference between two groups at 6w (P < 0.05).The BBB scores showed a significant difference at 4 w (P > 0.05) while the average area of myelin didn' t show the same trend as the Basso-Beattie-Bresnahan (BBB) scores between two groups.Conclusion Oscillating field stimulation can improve oligodendrocyte precursor cells proliferation in the injured spinal cord,which would contribute to the remyelination and neurological improvement after spinal cord injury.
