目的 观察鞘内注射普瑞巴林复合吗啡对大鼠骨癌痛的镇痛作用.方法 制备骨转移疼痛模型大鼠20只,随机分为4组(n=5):A组为鞘内注射生理盐水(NS)20 μl;B组为鞘内注射普瑞巴林(PGB) 30 μg;C组为鞘内注射吗啡(Mor)2 μg;D组为鞘内注射普瑞巴林(PGB) 30 μg+吗啡(Mor)2 μg.比较造模、给药后3、7、10d时的大鼠疼痛阈变化.结果 各时间点B组大鼠机械缩爪反射阈值(MWT) [3 d:25.1 g;7 d:25.6 g;10 d:24.9 g]、热刺激缩爪反应潜伏期(TWL) [3 d:(5.3±0.2) s;7 d:(5.2±0.8)s;10 d(5.2±0.4)s],均较A组明显增加(P<0.05);各时间点C组大鼠MWT[3 d:27.8 g;7 d:28.1 g;10 d:28.7 g]、TWL[3 d:(5.8±0.5) s;7 d:(5.9±0.7) s;10d(5.8±0.4)s],均较A组明显增加(P<0.05);各时间点D组大鼠MWT[3 d:39.8 g;7 d:40.2 g;10 d:41.1g]、TWL[3d:(9.8±0.7) s;7d:(9.7±0.2) s;10d(10.0±0.2)s],均较B、C组明显增加(P<0.05).结论 鞘内注射普瑞巴林复合吗啡可提高骨癌痛大鼠的MWT、TWL,对骨癌痛大鼠有较好的镇痛作用.

목적 관찰초내주사보서파림복합마배대대서골암통적진통작용.방법 제비골전이동통모형대서20지,수궤분위4조(n=5):A조위초내주사생리염수(NS)20 μl;B조위초내주사보서파림(PGB) 30 μg;C조위초내주사마배(Mor)2 μg;D조위초내주사보서파림(PGB) 30 μg+마배(Mor)2 μg.비교조모、급약후3、7、10d시적대서동통역변화.결과 각시간점B조대서궤계축조반사역치(MWT) [3 d:25.1 g;7 d:25.6 g;10 d:24.9 g]、열자격축조반응잠복기(TWL) [3 d:(5.3±0.2) s;7 d:(5.2±0.8)s;10 d(5.2±0.4)s],균교A조명현증가(P<0.05);각시간점C조대서MWT[3 d:27.8 g;7 d:28.1 g;10 d:28.7 g]、TWL[3 d:(5.8±0.5) s;7 d:(5.9±0.7) s;10d(5.8±0.4)s],균교A조명현증가(P<0.05);각시간점D조대서MWT[3 d:39.8 g;7 d:40.2 g;10 d:41.1g]、TWL[3d:(9.8±0.7) s;7d:(9.7±0.2) s;10d(10.0±0.2)s],균교B、C조명현증가(P<0.05).결론 초내주사보서파림복합마배가제고골암통대서적MWT、TWL,대골암통대서유교호적진통작용.
Objective To evaluate the analgesic effect of intrathecal injection of pregabalin and/or morphine on bone cancer pain in rats.Methods Twenty rats were used to establish the models of bone cancer pain,and randomly divided into 4 groups (n =5 each):group A (20 μl normal saline was administered intrathecally) ; group B (30 μg pregabalin was administered intrathecally) ; group C (2 μg morphine was administered intrathecally) ; group D (30 μg pregabalin +2 μg morphine).The pain threshold at different time points (3rd,7th and 10th day after dosing) was measured.Results In group B,mechanical paw withdrawal reflex threshold (MWT) at each time point [3 d:25.1 g; 7 d:25.6 g; 10 d:24.9 g],and thermal withdrawal latency (TWL) [3 d:(5.3 ±0.2) s; 7 d:(5.2 ±0.8) s; 10 d:(5.2 ±-0.4) s] were significantly higher than in group A (P < 0.05).In group C,MWT at each time point [3 d:27.8 g; 7 d:28.1 g; 10 d:28.7g],and TWL [3d:(5.8±0.5) s; 7 d:(5.9±0.7) s; 10 d:(5.8±0.4) s] were significantly higher than in groupA (P<0.05).In group D,MWT [3 d:39.8 g;7d:40.2 g; 10d:41.1 g],and TWL [3 d:(9.8 ±0.7) s; 7 d:(9.7 ±0.2) s; 10 d:(10.0 ±0.2) s] were singnificantly higher than in groups B and C (P < 0.05).Conclusion Pregabalin and/or morphine administrered intrathecally can raise the MWT and TWL,and produce a better analgesic effect in rat model of bone cancer pain.
