目的 应用蛋白组学技术检测出有助于下肢动脉硬化闭塞症(LASO)疾病诊断和治疗的生物标志物.方法 收集23例LASO患者和23例健康志愿者的血清,应用同位素标记相对和绝对定量(iTRAQ)结合二维液相色谱串联质谱(2D LC-MS/MS)技术鉴定LASO患者和正常人的差异表达蛋白,并利用Western blot法进行验证.结果 LASO患者与正常人比较,检测出20种表达量差异2倍以上的蛋白质,其中表达上调的蛋白有12种,下调的有8种.应用Westerm blot法验证高表达的嗅质蛋白-1(OLFM1),LASO患者血清中其表达量要明显高于对照组(2.05±0.66比0.96±0.18,P<0.01).结论 利用iTRAQ技术鉴定出LASO组与正常组的差异蛋白,可能成为诊断LASO的特异性血清生物标记物,并作为潜在的治疗靶点.

Objective Using a proteomic approach we aimed to determine potential serum biomarkers for diagnosis and managememt of lower limb arteriosclerosis obliterans (LASO).Methods Serum samples were collected from 23 patients with LASO and 23 healthy volunteer subjects.Isobaric tags for relative and absolute quantitation (iTRAQ)-tagging combined with two-dimensional liquid chromatographytandem masss pectrometry (2D LC-MS/MS) analysis was used to identification of differentially expressed proteins between LASO patients and healthy subjects.Candidates were validated in independent cohorts of LASO patients and healthy subjects by Westerm blotting.Results Using a proteomic approach,we identified 20 serum proteins are differentially expressed in LASO patients as compared with that in healthy subjects,of which 12 proteins increased more than two-fold,while 8 proteins decreased more than two-fold among the LASO patients.Western blotting test verified that Olfactomedin-1 (OLFM1) was up-regulated in LASO group as compared with the control group (2.05 ±0.66 vs 0.96 ±0.18,P <0.01).Conclusion Using an iTRAQ MS approach,we identification of differentially expressed proteins between LASO patients and healthy subjects,which might serve as potential candidates for diagnostic or therapeutic measures.
