目的 探讨高迁移率族蛋白B1(HMGB1)在乳腺癌中的表达及意义.方法 收集手术切除的20例乳腺癌患者的乳腺癌组织及其癌旁组织标本,采用逆转录-聚合酶链反应(RT-PCR)方法检测HMGB1基因的表达.从本院2012年1月至2012年12月手术的乳腺癌数据库中按时间顺序连续选择100例不同分期的乳腺癌组织及癌旁组织蜡块标本,采用免疫组织化学法检测HMGB1蛋白的表达,分析HMGB1表达与乳腺癌临床病理特征之间的关系.结果 与癌旁组织比较,HMGB1在乳腺癌组织中的基因表达量及蛋白表达率均较高,分别15.94±1.42比1.29±0.41、66%比18%(P<0.01);HMGB1蛋白表达程度与淋巴结转移和组织学分级明显相关(P<0.05),而与年龄、恶性肿瘤家族史及肿瘤直径无明显相关(P>0.05).结论 检测HMGB1基因及蛋白水平表达有助于乳腺癌的早期诊断及预后分析.

Objective To investigate the high mobility group protein B1 (HMGB1) expression and significance in breast cancer.Methods (1) Collecting breast cancer tissues and paracancerous tissues from 20 breast cancer cases who underwent surgery in January 2014 in the First Affiliated Hospital of Fujian Medical University,using reverse transcriptase-polymerase chain reaction (RT-PCR) methods to detect the expression of HMGB1 gene.(2) Selecting breast cancer tissue and paracancerous tissue paraffin block specimens from 100 patients with different breast cancer stages in The Breast Cancer Database of the hospital from January 2012 to December 2012 according to the time sequence,and using immunohistochemistry methods to detect the expression of HMGB1 protein,then analyzing the relationship between the expression of HMGB1 and clinical pathological characteristics of breast cancer.Results Compared to paracancerous tissues,HMGB1 ' s gene expression rate and the amount of protein expression are both higher in breast cancer tissues,respectively,15.94 ± 1.42 vs.1.29 ± 0.41,and 66% vs.18% (P < 0.01) ; the expression of HMGB1 protein is closely related to the extent of lymph node metastasis and tissue grading (P <0.05),but has no relation with age,family history of malignant tumor or tumor diameter (P >0.05).Conclusion The abnormal expression of HMGB1 gene and protein level have important guiding significance for early diagnosis and analysis on prognosis of breast cancer,and it may has a good application prospect to develop treatment on its target spot.
