目的 筛选并鉴定胃癌相关长链非编码RNA(LncRNA)在胃癌组织中的表达.方法 利用基因芯片筛选出胃癌组织及配对癌旁组织表达差异的LncRNA,采用实时定量逆转录聚合酶链反应(RT-qPCR)检测目标LncRNA在61例胃癌手术标本中的表达,结合病理特征探讨目标LncRNA的临床意义.结果 LncRNA AK054978在81.97%(50/61)样本中表达上调(P<0.01),上调水平与胃癌大小(P<0.01)、组织分级(P<0.01)、浸润深度(P<0.01)、淋巴结转移(P<0.01)、TNM分期(P<0.01)、脉管神经浸润(P<0.01)等临床病理特征呈正相关.结论 胃癌组织中LncRNAAK054978的高表达可能与胃癌的发生、发展、侵袭、转移相关.

Objective To screen and identify the existence of long non-coding RNA (LncRNAs) which are associated with gastric cancer,we investigate the expression of LncRNAs in gastric cancer tissues.Methods Gene microarray was used to detect the difference of expression profile of LncRNAs between gastric cancer tissues and matched adjacent non-tumorous tissues.Real-time quantitative reverse transcriptase-polymerase chain reaction (RT-qPCR) was used to verify the real expression levels of objective LncRNA in 61 surgically resected specimens.Combined with clinicopathological data,clinical significance of objective LncRNAs was clarified.Results The expression of AK054978 was up-regulated in 50 of 61 (81.97%) cases (P < 0.01) and associated with clinicopathological factors such as tumor size (P < 0.01),histologic grade (P < 0.01),depth of invasion (P < 0.01),lymphatic metastasis (P < 0.01),TNM stage (P < 0.01) and vessel or nerve invasion (P < 0.01).Conclusion LncRNA AK054978 may play an important role in the development,progression,invasion and metastasis of gastric cancer.
