目的 选择新生1d大鼠表皮干细胞(ESCs)接种在胶原修饰几丁质膜(C-CBM),构建具有生物活性的覆盖物,评估其修复效果及应用前景.方法 将16只裸鼠随机分4组(n=4):Ⅰ型胶原组(A组)、几丁质膜组(B组)、几丁质膜+ ESCs组(C组)和Ⅰ型胶原修饰几丁质膜+ESCs组(D组),制备裸鼠背部全层皮肤缺损模型,术后定期取材,组织包埋切片,苏木素-伊红(HE)染色,免疫组织化学检测CD34和CD200表达,观察创面早期滤泡性毛囊增生情况.结果 6周创面愈合皮肤以D组修复较好,肉眼观察创面较厚、红润,有皮纹;A组和B组的修复则较薄、呈淡紫色,易出血;C组愈合较慢;并发现表皮巢形成有规律地增多,D组>C组>B组>A组;D组CD34和CD200在手术后3d开始有毛囊干细胞增生,CD34表达延续到第4周,CD200表达延续到第6周,而C组CD34出现在第4周后,CD200出现在7d,D组是C组的2倍,A组和B组新生毛囊干细胞较少.结论 ESCs在C-CBM上生长良好,细胞可利用胶原的天然材料的黏附性,充分扩展,并参与创面修复,这些结果显示仿生膜有可能成为诱导性生物材料.
目的 選擇新生1d大鼠錶皮榦細胞(ESCs)接種在膠原脩飾幾丁質膜(C-CBM),構建具有生物活性的覆蓋物,評估其脩複效果及應用前景.方法 將16隻裸鼠隨機分4組(n=4):Ⅰ型膠原組(A組)、幾丁質膜組(B組)、幾丁質膜+ ESCs組(C組)和Ⅰ型膠原脩飾幾丁質膜+ESCs組(D組),製備裸鼠揹部全層皮膚缺損模型,術後定期取材,組織包埋切片,囌木素-伊紅(HE)染色,免疫組織化學檢測CD34和CD200錶達,觀察創麵早期濾泡性毛囊增生情況.結果 6週創麵愈閤皮膚以D組脩複較好,肉眼觀察創麵較厚、紅潤,有皮紋;A組和B組的脩複則較薄、呈淡紫色,易齣血;C組愈閤較慢;併髮現錶皮巢形成有規律地增多,D組>C組>B組>A組;D組CD34和CD200在手術後3d開始有毛囊榦細胞增生,CD34錶達延續到第4週,CD200錶達延續到第6週,而C組CD34齣現在第4週後,CD200齣現在7d,D組是C組的2倍,A組和B組新生毛囊榦細胞較少.結論 ESCs在C-CBM上生長良好,細胞可利用膠原的天然材料的黏附性,充分擴展,併參與創麵脩複,這些結果顯示倣生膜有可能成為誘導性生物材料.
목적 선택신생1d대서표피간세포(ESCs)접충재효원수식궤정질막(C-CBM),구건구유생물활성적복개물,평고기수복효과급응용전경.방법 장16지라서수궤분4조(n=4):Ⅰ형효원조(A조)、궤정질막조(B조)、궤정질막+ ESCs조(C조)화Ⅰ형효원수식궤정질막+ESCs조(D조),제비라서배부전층피부결손모형,술후정기취재,조직포매절편,소목소-이홍(HE)염색,면역조직화학검측CD34화CD200표체,관찰창면조기려포성모낭증생정황.결과 6주창면유합피부이D조수복교호,육안관찰창면교후、홍윤,유피문;A조화B조적수복칙교박、정담자색,역출혈;C조유합교만;병발현표피소형성유규률지증다,D조>C조>B조>A조;D조CD34화CD200재수술후3d개시유모낭간세포증생,CD34표체연속도제4주,CD200표체연속도제6주,이C조CD34출현재제4주후,CD200출현재7d,D조시C조적2배,A조화B조신생모낭간세포교소.결론 ESCs재C-CBM상생장량호,세포가이용효원적천연재료적점부성,충분확전,병삼여창면수복,저사결과현시방생막유가능성위유도성생물재료.
Objective To select scaffold as a covering with bio-inspired which from epidermal stem cells (ESCs) of the newborn day 1 rats and cultured on chitin biomimetic inembrane (CBM) or collagen-modified chitin biomimetic membrane (C-CBM).Evaluate the effect of the biomimetic membrane repair and its application prospect.Methods sixteen nude mice were randomly divided into four groups (n =4 each).Type Ⅰ collagen group (A),chitin biomimetic group (B),chitin biomimetic group plus ESCs group (C) and Type Ⅰ collagen modified chitin biomimetic membrane group (D).Full-thickness skin defects model was establish on the backside of the nude mice.The samples analyzed after operation by embedding slice and hematoxylin and eosin (HE) staining,expression of the CD34 and CD200 by Immunohistochemistry.Observe the proliferation of the follicular hair follicles in early stage.Results Group D showed the repaired wound skin was good for repair which is thick and red with re-epithelialization in 6 weeks.While group A & B showed the repaired wound skin was relatively thin,pale purple,bleeding easily.Group C was re-epithelialization slowly.Further found that the skin nest formed the regular increase in repair skin tissue.Group D > C > B > A ; CD34 and CD200 showed proliferation of the hair follicle stem cells post surgery d3 in group D.Expression CD34 positive extend for 4 weeks and CD200 positive extend for 6 weeks.Group C was expression CD34 positive in 4 weeks and CD200 start in 7 d.Group D was 2 times higher than group C.Less in renascence hair follicle stem cells of the group A and B.Conclusion ESCs grow up well in collagen-modified chitin biomimetic membrane.To take advantage adhesion of natural materials and cell extend abundant.Furthermore participate in wound healing.These data shows bio-inspired membrane can be applied inductivity biological materials.
