目的 观察小分子抑制剂Spautin-1对骨肉瘤细胞株(U2OS)化疗敏感性的影响.方法 培养U2OS细胞,使用有效自噬抑制浓度的Spautin-1(10 μmol/L)和顺铂(5μmol/L)处理U2OS,细胞计数试剂盒(CCK-8)法检测Spautin-1对于顺铂(Cisplatin)介导的U2OS的增殖抑制作用的影响,流式细胞术和Western blot检测Spautin-1对顺铂介导的U2OS的凋亡诱导作用的影响.结果 有效自噬抑制浓度的Spautin-1(10 μmol/L)对U2OS无明显的生长抑制及凋亡诱导作用(P>0.05),而此浓度的Spautin-1可明显增强顺铂对U2OS的增殖抑制(P<0.05),与顺铂处理组(16.5%)比较,Spautin-1与顺铂共处理后U2OS细胞凋亡比例明显增加(35.0%),切割后的半胱氨酰天冬氨酸特异性蛋白酶-3(Caspase-3)和聚腺苷二磷酸核糖聚合酶(PARP)明显升高.结论 小分子抑制剂Spautin-1可增强U2OS对顺铂的敏感性.

Objective To study the effect of autophagy inhibitor Spautin-1 on osteosarcoma cell sensitivity to cisplatin-induced cell proliferation inhibition and apoptosis.Methods Osteosarcoma cell line U2OS cells were treated with an effective inhibition concentration of Spautin-1 (10 μmol/L).Cell proliferation was measured by cell counting kit-8 (CCK-8),and cell apoptosis was measured by flow cytometry and Western blotting.Results Spautin-1 has no obvious effect on U2OS cell proliferation and apoptosis (P > 0.05).However,Spautin-1 sensitized osteosarcoma cells to cisplatin-induced cell proliferation inhibition and apoptosis.The flow cytometry indicated that the apoptosis rate in Spautin-1 + cisplatin group (35.0%) was significantly higher than that in cisplatin group (16.5%).The cleaved Caspase-3 and poly adenosine diphosphate-ribose polymerase (PARP) were also increased in Spautin-1 +cisplating group as compared with cisplatin group.Conclusion The autophagy inhibitor Spautin-1 sensitizes osteosarcoma cells to cisplatin-induced cell proliferation inhibition and apoptosis.
