目的 比较神经生长因子(NGF)与尼莫地平两种药物对大鼠糖尿病周围神经病变(DPN)预处理的影响.方法 用链脲佐菌素(STZ)诱导建立糖尿病(DM)大鼠模型并随机分组:Ⅰ:空白对照组(Control);Ⅱ:糖尿病组(DM);Ⅲ:糖尿病+β-神经生长因子组(DM+ β-NGF);Ⅳ:糖尿病+尼莫地平组(DM+ Nimodipine).检测各组坐骨神经运动传导速度(MNCV)以及坐骨神经病理改变.结果 坐骨神经MNCV的测量结果:8周时,Control组[(50.70±1.12) m/s]与DM组[(45.80±1.12) m/s]比较,差异有统计学意义(P<0.05),β-NGF组[(49.50±1.08) m/s]、尼莫地平组[(48.50±0.81) m/s]分别与DM组[(45.80±1.12) m/s]比较,差异有统计学意义(P<0.05);12周时,β-NGF组[(49.50±1.08) m/s]、尼莫地平组[(48.50±0.81) m/s]分别与Control组[(50.90±1.31) m/s]比较,差异有统计学意义(P<0.05),β-NGF组[(49.50±1.08) m/s]与尼莫地平组[(48.50±0.81) m/s]比较,差异有统计学意义(P<0.05).与Control比较,其余3组坐骨神经形态学结构均有不同程度的破坏,DM组>尼莫地平组>β-NGF组.结论 NGF和尼莫地平均对大鼠坐骨神经糖尿病周围神经病变有预保护作用,但不能阻止其发生和发展,NGF对其预保护作用要优于尼莫地平,并随时间延长效果也逐渐明显.

Objective The present study was planned to compare the nerve growth factor (NGF)and nimodipine on diabetic peripheral neuropathy (DPN) the effects of pretreatment.Methods The diabetic rats model that induced by streptozotocin (STZ) were randomly divided into groups:Ⅰ:blank group (Control) ; Ⅱ:diabetes mellitus group (DM) ; Ⅲ:diabetes + β-NGF group (DM + β-NGF) ; Ⅳ:diabetes + nimodipine group (DM + Nimodipine).The level of sciatic morter nerve conduction velocity and pathology of the animals were checked.Results At 8 week,compared with control group [(50.70 ±1.12) m/s],the MNCV of DM group [(45.80 ± 1.12) m/s] decreased significantly (P < 0.05).β-NGF group [(49.50 ± 1.08) m/s] and nimodipine group [(48.50 ±0.81) m/s] compared with the DM group [(45.80 ± 1.12) m/s] was statistically significant (P < 0.05) ; At 12 week,compared with Control group [(43.90 ± 1.03) m/s],β-NGF group [(49.50 ± 1.08) m/s] and Nimodipine group [(48.50 ± 0.81) m/s] was statistically significant (P < 0.05),Compared with Nimodipine group [(48.50 ±0.81) m/s],β-NGF group [(49.50 ± 1.08) m/s] has better protection to the nerve and there is obviously difference (P < 0.05).Compared with Control group,the structure of the sciatic nerve was damaged in different degree.β-NGF group show little damage than nimodipine group,and the sciatic structure of DM group is the worst.Conclusion Nerve growth factor and nimodipine has protective effect on sciatic nerve in diabetic rats,but cannot prevent the occurrence and development.Protective effect of NGF on its superior to Nimodipine,and extended the effects become apparent over time.
