目的 探讨硼替佐米联合丙戊酸钠对人胰腺癌细胞株SW1990的协同凋亡作用及其机制.方法 以浓度为100 nmol/L的硼替佐米联合3 mmol/L的丙戊酸钠干预SW1990细胞株,采用噻唑蓝(MTT)法检测细胞增殖;膜联蛋白V(Annexin V)/碘化丙锭(PI)双染法检测细胞早期凋亡;反转录-聚合酶链反应(RT-PCR)检测细胞凋亡因子生存素(Survivin)、半胱氨酰天冬氨酸特异性蛋白酶(Caspase)-3的mRNA表达水平;Western blot法检测Survivin、Caspase-3前体(pro-Caspase-3)、裂解的Caspase-3(cleaved-Caspase-3)蛋白表达水平.结果 硼替佐米单药对SW1990的生长抑制作用较弱,而丙戊酸钠单药可有效抑制SW1990细胞的增殖,硼替佐米单药组干预SW1990细胞48 h的早期凋亡率为(13.47 ±2.21)%,而丙戊酸钠单药组为(26.73±2.36)%,两药联合组为(47.06±2.76)%.硼替佐米单药组Survivin mRNA及蛋白水平表达无明显改变、而丙戊酸钠单药组及两药联合组的Survivin、Caspase mRNA及蛋白表达水平均明显下调,cleaved-Caspase-3蛋白表达水平明显上调,各项检测指标联合用药组与单药组比较差异均有统计学意义(P<0.05),且两药联合有协同作用,可提高SW1990对硼替佐米的敏感性.结论 硼替佐米联合丙戊酸钠对诱导人胰腺癌细胞株SW1990凋亡有协同增效作用,其机制可能与下调Survivin的表达水平相关.

목적 탐토붕체좌미연합병무산납대인이선암세포주SW1990적협동조망작용급기궤제.방법 이농도위100 nmol/L적붕체좌미연합3 mmol/L적병무산납간예SW1990세포주,채용새서람(MTT)법검측세포증식;막련단백V(Annexin V)/전화병정(PI)쌍염법검측세포조기조망;반전록-취합매련반응(RT-PCR)검측세포조망인자생존소(Survivin)、반광안선천동안산특이성단백매(Caspase)-3적mRNA표체수평;Western blot법검측Survivin、Caspase-3전체(pro-Caspase-3)、렬해적Caspase-3(cleaved-Caspase-3)단백표체수평.결과 붕체좌미단약대SW1990적생장억제작용교약,이병무산납단약가유효억제SW1990세포적증식,붕체좌미단약조간예SW1990세포48 h적조기조망솔위(13.47 ±2.21)%,이병무산납단약조위(26.73±2.36)%,량약연합조위(47.06±2.76)%.붕체좌미단약조Survivin mRNA급단백수평표체무명현개변、이병무산납단약조급량약연합조적Survivin、Caspase mRNA급단백표체수평균명현하조,cleaved-Caspase-3단백표체수평명현상조,각항검측지표연합용약조여단약조비교차이균유통계학의의(P<0.05),차량약연합유협동작용,가제고SW1990대붕체좌미적민감성.결론 붕체좌미연합병무산납대유도인이선암세포주SW1990조망유협동증효작용,기궤제가능여하조Survivin적표체수평상관.
Objective To investigate the effect and mechanism of bortezomib in combination with valproate on human pancreatic cancer cell line SW1990.Methods Bortezomib,valproate were tested alone and in combination for their abilities to inhibit SW1990 cell line proliferation by methyl thiazol tetrazolium (MTT).The early apoptosis of SW1990 cell line were detected by Annexin V/propidium iodide (PI)double staining method,and the expression of Survivin and Caspase-3 mRNA were detected by reverse transcriptase-polymerase chain reaction (RT-PCR).The content of Survivin and pro-Caspase-3 and cleaved-Caspase-3 were evaluated by Western blotting.Results Bortezomib could inhibited faintly the proliferation of SW1990 cell line,however,valproate and combination could inhibited effectively the proliferation of SW1990 cell line.The early stage apoptosis rate of SW1990 cells was (13.47 ±2.21)% when treated with bortezomib,and was (26.73 ± 2.36) % when treated with valproate,and was (47.06 ±2.76)% when they were treated with bortezomib combined with valproate. The expression of Survivin mRNA and proteins had no significant changes in bortezomib group.The expression of Survivin,Caspase mRNA and proteins levels were down-regulated in valproate and combination group.Comparisons of treated groups and control group have significant difference (P < 0.05) and comparisons of combination groups and single drug group have also significant difference (P < 0.05).Conclusion Bortezomib interacts synergistically with valproate to induce apoptosis in SW1990 cell line.The potential mechanism is down-regulated Survivin mRNA and protein levels to promote apoptosis of cells.
