目的 探讨中药首乌益智胶囊对血管性认知障碍模型大鼠脑组织海马神经元超微结构的影响.方法 血管性认知障碍大鼠模型制作采用右侧大脑中动脉栓塞再灌注法,造模成功后将大鼠随机分为首乌益智组(给予首乌益智胶囊混悬液灌胃,5.18 g/100 g)、脑复康组(给予脑复康混悬液灌胃,3.24 g/100 g)、模型组(给予0.9%氯化钠注射液灌胃,1 ml/100 g),每组10只,另设假手术组10只,共40只大鼠.药物干预28 d后,采用透镜电镜观察各组大鼠海马区神经元的超微结构.结果 假手术组10只大鼠海马神经元细胞核清晰可辨,核内细胞器丰富且结构清晰;模型组9只大鼠海马锥体细胞明显缺失、线粒体肿胀明显、血管周围水肿;脑复康组7只大鼠海马区神经细胞核膜结构较清晰,部分线粒体絮状变,髓鞘结构较清晰,与模型组相比明显改善;首乌益智胶囊组9只大鼠细胞器基本正常.结论 首乌益智胶囊能明显改善VCI大鼠神经元的损伤.
目的 探討中藥首烏益智膠囊對血管性認知障礙模型大鼠腦組織海馬神經元超微結構的影響.方法 血管性認知障礙大鼠模型製作採用右側大腦中動脈栓塞再灌註法,造模成功後將大鼠隨機分為首烏益智組(給予首烏益智膠囊混懸液灌胃,5.18 g/100 g)、腦複康組(給予腦複康混懸液灌胃,3.24 g/100 g)、模型組(給予0.9%氯化鈉註射液灌胃,1 ml/100 g),每組10隻,另設假手術組10隻,共40隻大鼠.藥物榦預28 d後,採用透鏡電鏡觀察各組大鼠海馬區神經元的超微結構.結果 假手術組10隻大鼠海馬神經元細胞覈清晰可辨,覈內細胞器豐富且結構清晰;模型組9隻大鼠海馬錐體細胞明顯缺失、線粒體腫脹明顯、血管週圍水腫;腦複康組7隻大鼠海馬區神經細胞覈膜結構較清晰,部分線粒體絮狀變,髓鞘結構較清晰,與模型組相比明顯改善;首烏益智膠囊組9隻大鼠細胞器基本正常.結論 首烏益智膠囊能明顯改善VCI大鼠神經元的損傷.
목적 탐토중약수오익지효낭대혈관성인지장애모형대서뇌조직해마신경원초미결구적영향.방법 혈관성인지장애대서모형제작채용우측대뇌중동맥전새재관주법,조모성공후장대서수궤분위수오익지조(급여수오익지효낭혼현액관위,5.18 g/100 g)、뇌복강조(급여뇌복강혼현액관위,3.24 g/100 g)、모형조(급여0.9%록화납주사액관위,1 ml/100 g),매조10지,령설가수술조10지,공40지대서.약물간예28 d후,채용투경전경관찰각조대서해마구신경원적초미결구.결과 가수술조10지대서해마신경원세포핵청석가변,핵내세포기봉부차결구청석;모형조9지대서해마추체세포명현결실、선립체종창명현、혈관주위수종;뇌복강조7지대서해마구신경세포핵막결구교청석,부분선립체서상변,수초결구교청석,여모형조상비명현개선;수오익지효낭조9지대서세포기기본정상.결론 수오익지효낭능명현개선VCI대서신경원적손상.
Objective To investigate the effect of Shouwuyizhi (SWYZ) capsule on the ultratructure of brain tissue in experimental rats with vascular cognitive impairment (VCI).Methods The VCI model rats were established with MCAO method.All experimental rats were divided into Shouwuyizhi group,Naofukang group,model group and sham operation group.After 4 weeks,the rats cerebral histomorphological change was observed.Results In the sham operation group,the nucleus of hippocampal neurons was clear,organelles were abundant and had clear nuclei structure in 10 rats.In the model group,hippocampal pyramidal cells were missed with swollen mitochondria and perivascular edema in 9 rats.In the Naofukang group,hippocampus nerve cells' membrane structure was relatively clear; some mitochondria presented flocculent degeneration and myelin structure was clear in 7 rats.In the Shouwuyizhi group,the loss of pyramidal cells was reduced; organelles were basically normal; perivascular edema was significantly relieved in 9 rats.Conclusion SWYZ capsule can significantly improve the VCI rat neuronal injury.
