背景与目的:内质网分子伴侣29(endoplasmic reticulum molecular chaperons 29,ERp29)是一种内质网应激蛋白,其高表达与肿瘤转移相关。本研究旨在探讨ERp29在小鼠腹水型肝癌高、低淋巴道转移株中的表达及与肝癌淋巴道转移的关系。方法:构建小鼠腹水型肝癌细胞株(高转移株Hca-F、低转移株Hca-P)淋巴道转移动物模型,通过免疫组织化学、蛋白质印迹法(Western blot)和流式细胞仪等方法检测ERp29在Hca-F、Hca-P细胞株中的表达情况。结果:免疫组织化学检测结果显示,ERp29在Hca-F、Hca-P中均呈阳性表达,定位于细胞质,部分细胞核也有表达。Western blot检测结果显示,ERp29在Hca-F、Hca-P中的表达吸光度值分别为0.97±0.03和1.17±0.03,ERp29在Hca-F细胞株中的表达(吸光度值)明显低于Hca-P,差异有统计学意义(P<0.01)。流式细胞仪检测结果显示,ERp29在Hca-F细胞株中的相对荧光强度值(375.27±47.33)显著低于Hca-P(623.91±46.80),差异有统计学意义(P<0.01)。结论:ERp29在小鼠腹水型肝癌细胞株Hca-F和Hca-P均有表达,且在Hca-F中表达较低,而在Hca-P中表达较高。ERp29可能与肝癌淋巴道转移密切相关。

배경여목적:내질망분자반려29(endoplasmic reticulum molecular chaperons 29,ERp29)시일충내질망응격단백,기고표체여종류전이상관。본연구지재탐토ERp29재소서복수형간암고、저림파도전이주중적표체급여간암림파도전이적관계。방법:구건소서복수형간암세포주(고전이주Hca-F、저전이주Hca-P)림파도전이동물모형,통과면역조직화학、단백질인적법(Western blot)화류식세포의등방법검측ERp29재Hca-F、Hca-P세포주중적표체정황。결과:면역조직화학검측결과현시,ERp29재Hca-F、Hca-P중균정양성표체,정위우세포질,부분세포핵야유표체。Western blot검측결과현시,ERp29재Hca-F、Hca-P중적표체흡광도치분별위0.97±0.03화1.17±0.03,ERp29재Hca-F세포주중적표체(흡광도치)명현저우Hca-P,차이유통계학의의(P<0.01)。류식세포의검측결과현시,ERp29재Hca-F세포주중적상대형광강도치(375.27±47.33)현저저우Hca-P(623.91±46.80),차이유통계학의의(P<0.01)。결론:ERp29재소서복수형간암세포주Hca-F화Hca-P균유표체,차재Hca-F중표체교저,이재Hca-P중표체교고。ERp29가능여간암림파도전이밀절상관。
Background and purpose:ERp29 belongs to endoplasmic reticulum stress proteins and might play roles in neoplasm metastasis. This study aimed to investigate the expression of ERp29 in Hca-F and Hca-P cells and to elucidate its role in lymphatic metastasis.Methods:Immunohistochemistry, Western blot and lfow cytometry analysis were used to detect the expression of ERp29 in Hca-F and Hca-P.Results:The results of immunohistochemistry suggested that ERp29 protein was located at cytoplasm of hepatic cells and some were also detected in the nucleus. The results of western blot suggested that ERp29 had positive expression in Hca-F and Hca-P cells. Its expression in Hca-F cells was apparently lower than that in Hca-P cells. And there was statically different between Hca-F and Hca-P cells (P<0.01). The relative lfuorescence intensity of ERp29 protein was signiifcantly lower in Hca-F cells (375.27±47.33) than that in Hca-F cells (623.91±46.80) by lfow cytometry (P<0.01).Conclusion:The different expression of ERp29 may be related to the potential ability of tumor lymphatic metastasis in Hca-F and Hca-P cells.
