目的:建立款冬花总生物碱含量测定方法,并对款冬花生品和炮制品的含量进行比较研究。方法:采用酸性染料比色法,检测波长:417.5 nm。结果:款冬花总生物碱在6.75~135.00μg范围内呈良好的线性关系,回归方程:y=85.228x-0.0401(r=0.9995)。款冬花生品样品中总生物碱平均含量为0.1070 mg·g-1,蜜炙品样品中总生物碱平均含量为0.1633 mg·g-1,甘草炙品样品中总生物碱平均含量为0.0861 mg·g-1。结论:该方法经方法学验证可用于款冬花中总生物碱含量的测定。款冬花生品经炮制后,总生物碱含量发生变化,且炮制方法不同,总生物碱含量变化不同。蜜炙品的总生物碱含量最高,生品次之,甘草炙品最低。
目的:建立款鼕花總生物堿含量測定方法,併對款鼕花生品和砲製品的含量進行比較研究。方法:採用痠性染料比色法,檢測波長:417.5 nm。結果:款鼕花總生物堿在6.75~135.00μg範圍內呈良好的線性關繫,迴歸方程:y=85.228x-0.0401(r=0.9995)。款鼕花生品樣品中總生物堿平均含量為0.1070 mg·g-1,蜜炙品樣品中總生物堿平均含量為0.1633 mg·g-1,甘草炙品樣品中總生物堿平均含量為0.0861 mg·g-1。結論:該方法經方法學驗證可用于款鼕花中總生物堿含量的測定。款鼕花生品經砲製後,總生物堿含量髮生變化,且砲製方法不同,總生物堿含量變化不同。蜜炙品的總生物堿含量最高,生品次之,甘草炙品最低。

Objective:To establish the method of determination of total alkaloid content inTussilago farfara, and make a comparative study on total alkaloid content inTussilago farfarabefore and after processing. Methods:The content of total alkaloids was determined by acid dye colorimetry, with UV detection wavelength set at 417.5 nm.Results:The total alkaloid content inTussilago farfara was in good linear in the range of 6.75-135.00 μg, and its regression equation wasy=85.228x-0.0401,r=0.9995. The total alkaloid content was 0.1070 mg·g-1 in crudeTussilago farfara, 0.1633 mg·g-1 inTussilago farfara after honey-fried processing, and 0.0861 mg·g-1 inTussilago farfara after liquorice juice processing.Conclusion:This methodologically validated method can be used for determination of the total alkaloids inTussilago farfara. The content of total alkaloids changed after processing, and it changed with the processing methods simultaneously. The total alkaloid content was the highest inTussilago farfara after honey-fried processing, medium in crudeTussilago farfara and the least after liquorice juice processing.
