目的:通过观察各实验组小鼠肺部炎症情况和检测支气管肺泡灌洗液( BALF)中细胞因子白介素-12和干扰素-γ( IFN-γ)的水平,探讨糖皮质激素对小鼠支气管哮喘的治疗作用机制。方法清洁级BALB/c小鼠随机分为正常组、哮喘组、布地奈德组。以卵清蛋白致敏激发法建立哮喘小鼠模型。显微镜下观察肺组织病理切片;收集BALF,计算嗜酸性粒细胞( EOS)绝对计数百分比;用ELISA法测定BALF中IL-12和IFN-γ的水平,并做相关性分析。结果哮喘组小鼠BALF中白细胞总数、EOS数目和百分比分别与正常组、布地奈德组相比均明显增加(P<0.01),在正常组、布地奈德组之间比较差异无显著性(P>0.05)。哮喘组小鼠BALF中IL-12和IFN-γ水平分别与正常组和布地奈德组相比均减低( P<0.05),在正常组和布地奈德组对这两个指标进行比较差异无显著性( P>0.05)。各组小鼠BALF中IL-12和IFN-γ水平都呈正相关关系。结论布地奈德可促进IL-12和IFN-γ的表达,减轻肺部炎症,可能是糖皮质激素治疗哮喘的重要机制。

목적:통과관찰각실험조소서폐부염증정황화검측지기관폐포관세액( BALF)중세포인자백개소-12화간우소-γ( IFN-γ)적수평,탐토당피질격소대소서지기관효천적치료작용궤제。방법청길급BALB/c소서수궤분위정상조、효천조、포지내덕조。이란청단백치민격발법건립효천소서모형。현미경하관찰폐조직병리절편;수집BALF,계산기산성립세포( EOS)절대계수백분비;용ELISA법측정BALF중IL-12화IFN-γ적수평,병주상관성분석。결과효천조소서BALF중백세포총수、EOS수목화백분비분별여정상조、포지내덕조상비균명현증가(P<0.01),재정상조、포지내덕조지간비교차이무현저성(P>0.05)。효천조소서BALF중IL-12화IFN-γ수평분별여정상조화포지내덕조상비균감저( P<0.05),재정상조화포지내덕조대저량개지표진행비교차이무현저성( P>0.05)。각조소서BALF중IL-12화IFN-γ수평도정정상관관계。결론포지내덕가촉진IL-12화IFN-γ적표체,감경폐부염증,가능시당피질격소치료효천적중요궤제。
[ ABSTRACT] Objective To explore the therapeutic mechanism of glucocorticoid that be used to cure the bronchial asthma in mice by observing the condition of lung inflammation and the levels of interleukin-12 and interferon-γ( IFN-γ) in bronchoalveolar lavage fluid ( BALF) of each experimental group .Methods Clean level BALB/c mice were randomly divided into normal group , asthma group and budesonide group .The mouse models of asthma were established by ovalbumin sensitization excitation method .The lung tissue pathological sections were observed under the microscope;BALF were collected to calculate the percentage of eosinophils (EOS) counts,the levels of IL-12 and IFN-γin BALF were measured by ELISA method and correlation analysis among them were made .Results Compared with the normal grope and budesonide group ,the total of WBC ,the counts of EOS and percentage in BALF of asthmatic grope were significantly increased ( P<0.01),however,between the normal group and budesonide group ,there were no statistically significance(P>0.05).Conclusion Budes-onide can promote the expression of IL-12 and IFN-γand reduce lung inflammation ,it may be the important mechanism of the glucocorticoid treatment of asthma .
