Lacidipine%Endothelin%Calreticulin%Rats of hypertensive model
目的:探讨拉西地平对肾性高血压模型大鼠血浆内皮素(ET)及动脉血管内质网应激相关因子钙网蛋白(CRT)表达的影响。方法选择健康雄性Wistar大鼠40只,7~8周龄,随机分为假手术组、模型组、拉西地平组、阳性对照组,每组10只。假手术组仅分离血管不行缩窄术,模型组、拉西地平组以及阳性对照组大鼠行肾动脉缩窄术。拉西地平组给予拉西地平(0.72 mg/kg/d),阳性对照组大鼠给予卡托普利(35 mg/kg/d),手术前1周开始药物干预,每天1次,持续4周,假手术组和模型组给予等量生理盐水。给药4周后,测量大鼠血压,计算平均动脉压;采用免疫组化法测定CRT表达水平;放免法测定血浆ET水平。结果与假手术组比较,模型组平均动脉压升高[(119.83±9.41)mmHg vs.(180.75±11.26) mmHg],差异有显著统计学意义(P<0.01)。与模型组比较,阳性对照组[(180.75±11.26)mmHg vs.(138.68±10.46)mmHg]和拉西地平组[(180.75±11.26)mmHg vs.(139.54±10.21)mmHg]平均动脉压均降低,差异有显著统计学意义(P均<0.01)。与假手术组比较,模型组CRT表达的AOD值[(0.125±0.04) vs.(0.436±0.05)]增加,差异有显著统计学意义(P<0.01)。与模型组比较,阳性对照组大鼠CRT的AOD值[(0.436±0.05) vs.(0.195±0.04)]降低,拉西地平组大鼠CRT的AOD值[(0.436±0.05) vs.(0.209±0.03)]也降低,差异有显著统计学意义(P均<0.01)。与假手术组比较,模型组ET表达的AOD值[(1.62±0.45) vs.(3.86±1.05)]增加,差异有统计学意义(P<0.05)。与模型组比较,阳性对照组大鼠ET的AOD值[(3.86±1.05)vs.(2.04±0.55)]降低,拉西地平组大鼠ET的AOD值[(3.86±1.05) vs.(2.15±0.84)]也降低,差异有统计学意义(P均<0.05)。结论拉西地平可降低肾性高血压大鼠平均动脉压,降低ET水平,可能通过下调内质网应激,降低CRT表达,保护血管平滑肌细胞。

Objective To investigate the influences of lacidipine on expressions of endothelin (ET) and calreticulin (CRT) of vascular smooth muscle cell (VSMC) in rats of renal hypertensive model. Methods Male Wistar rats (n=40, aged from 7 to 8 w) were randomly divided into sham-operation group, model group, lacidipine group and positive control group (each n=10). The sham-operation group was not given and other groups were given renal artery constriction. The lacidipine group was treated with lacidipine (0.72 mg/kg/d), and positive control group was given captopril (35 mg/kg/d) once a day for 4 w before surgery. The sham-operation group and model group were given normal saline water. After 4 w, blood pressure was detected and mean arterial pressure was calculated. The expression of CRT was detected by using immunohistochemistry technique, and level of plasma ET was detected by using radioimmunoassay (RIA). Results Compared with sham-operation group, mean arterial pressure increased [(119.83±9.41) mmHg vs. (180.75±11.26) mmHg] in model group (P<0.01). Compared with model group, mean arterial pressure decreased in positive control group [(180.75±11.26) mmHg vs. (138.68±10.46) mmHg] and lacidipine group [(180.75±11.26) mmHg vs. (139.54±10.21) mmHg, all P<0.01]. Compared with sham-operation group, AOD value of CRT expression increased [(0.125±0.04) vs. (0.436±0.05)] in model group (P<0.01). Compared with model group, AOD value of CRT expression decreased in positive control group [(0.436 ±0.05) vs. (0.195±0.04)] and also decreased in lacidipine group [(0.436±0.05) vs. (0.209±0.03), all P<0.01]. Compared with sham-operation group, AOD value of ET expression increased in model group [(1.62±0.45) vs. (3.86 ±1.05), P<0.05]. Compared with model group, AOD value of ET expression decreased in positive control group [(3.86±1.05) vs. (2.04±0.55)], and decreased also in lacidipine group [(3.86±1.05) vs. (2.15±0.84), all P<0.05]. Conclusion Lacidipine can reduce mean arterial pressure and ET level, decrease CRT expression and protect VSMC through down-regulating endoplasmic reticulum stress in rats with renal hypertension.
