研究多西环素对甲状腺癌SW579细胞增殖、凋亡和侵袭的影响。采用MTT技术检测细胞增殖能力的变化,荧光分光光度计检测线粒体膜电位的变化,Westernblot技术检测细胞色素C(CytC)和基质金属蛋白酶2(MMP‐2)蛋白表达,Transwell小室侵袭实验检测细胞侵袭能力的变化。结果显示,与对照组相比,多西环素组细胞存活率,线粒体膜电位,MMP‐2蛋白表达,侵袭细胞数均明显降低(P<0.05),CytC表达明显增高(P<0.05),呈浓度和时间依赖性(P<0.05)。因此,多西环素能有效抑制SW579细胞增殖和侵袭,并且能诱导其细胞凋亡。

To detect the effects of doxycycline on cell proliferation ,apoptosis ,and invasion as well as the metastasis of of thyroid squamous cell carcinoma cell line SW579 .The ability of cell proliferation in each group was detected using MTT assay ,the mitochondrial membrane potential was measured with fluorescence spectrophotometer ,the expression levels of cytochrome C (CytC) and MMP‐2 in each group were measured by western blot ,the ability of invasion in each group was measured with Transwell assays .The results showed that the cell survival rates ,invasion rate and the MMP‐2 expression of SW579 decreased dramatically (P<0 .05) ,while the expression of CytC increased significantly in the doxycycline group compared with control group (P< 0 .05) ,which are all in a concentration and time‐dependent manners (P< 0 .05) . Doxycycline can effectively inhibit the cell proliferation and invasion ,and induce apoptosis of SW579 cells .
