目的::观察雷公藤多苷( multiglucosides of triptergium wilfordii,GTW)对糖尿病肾病( diabetic nephropathy,DN)大鼠肾组织甘露糖结合凝集素(mannose-binding lectin,MBL)、白细胞介素6(interleukin 6,IL-6)、肿瘤坏死因子α(tissue necrosis factor-α,TNF-α)表达影响,探讨MBL、IL-6、TNF-α与DN关系,阐明GTW对DN保护作用及可能机制。方法:40只雄性SD大鼠随机分为实验组和对照组,实验组高糖高脂饮食6周后,以链脲佐菌素( streptozocin,STZ)建立DN大鼠模型。造模成功的大鼠随机分为DN组和GTW治疗组,治疗组经口灌胃GTW(10 mg·kg-1·d-1)8周。第14周末称体重并查24 h尿蛋白定量、血清生化指标、观察肾脏结构及炎细胞浸润程度,免疫组化法检测MBL-A、IL-6、TNF-α蛋白在肾脏中的表达,RT-PCR法检测肾组织MBL1mRNA表达情况。结果:GTW可减轻DN大鼠蛋白尿,降低血清肌酐、尿素氮水平,下调肾组织MBL、IL-6、TNF-α表达。结论:GTW对DN大鼠肾组织有保护作用,可能与GTW减少了DN大鼠肾组织MBL、IL-6、TNF-α表达,减轻了免疫炎症反应对肾脏的损伤有关。

목적::관찰뢰공등다감( multiglucosides of triptergium wilfordii,GTW)대당뇨병신병( diabetic nephropathy,DN)대서신조직감로당결합응집소(mannose-binding lectin,MBL)、백세포개소6(interleukin 6,IL-6)、종류배사인자α(tissue necrosis factor-α,TNF-α)표체영향,탐토MBL、IL-6、TNF-α여DN관계,천명GTW대DN보호작용급가능궤제。방법:40지웅성SD대서수궤분위실험조화대조조,실험조고당고지음식6주후,이련뇨좌균소( streptozocin,STZ)건립DN대서모형。조모성공적대서수궤분위DN조화GTW치료조,치료조경구관위GTW(10 mg·kg-1·d-1)8주。제14주말칭체중병사24 h뇨단백정량、혈청생화지표、관찰신장결구급염세포침윤정도,면역조화법검측MBL-A、IL-6、TNF-α단백재신장중적표체,RT-PCR법검측신조직MBL1mRNA표체정황。결과:GTW가감경DN대서단백뇨,강저혈청기항、뇨소담수평,하조신조직MBL、IL-6、TNF-α표체。결론:GTW대DN대서신조직유보호작용,가능여GTW감소료DN대서신조직MBL、IL-6、TNF-α표체,감경료면역염증반응대신장적손상유관。
Objective:To observe the effects of multiglucosides of triptergium wilfordii on the expression of Man-nose-Binding Lectin(MBL)、interleukin 6(IL-6)、tissue necrosis factor-α(TNF-α) in renal tissue of diabetic nephropathy rats, dis-cussing the relationship between MBL、IL-6、TNF-α and diabetic nephropathy. In order to explore the potential protective mecha-nism of multiglucosides of triptergium wilfordii on diabetic nephropathy. Methods:Forty male Sprague-Dawlay( SD) rats were ran-domly divided into experimental group and normal control group, after 6 weeks high-suger and high-fat diet, the experimental group diabetes were injected into streptozocin( STZ) for DN rat model. The Rat models were randomly divided into diabetes mellitus control group ( group DN) , and multiglucosides of triptergium wilfordii treated diabetes mellitus group( group GTW) , that were treated with multiglucosides of triptergium wilfordii (10 mg·kg-1 ·d-1 ) for 8 weeks. After 14 weeks, the weight of rat、24 hours protein(24 h U-TP)、biochemical indexes were measured. Partial nephridial tissues were treated with HE staining. Using immunohistochemistry to observe the protein expression of MBL-A、IL-6、TNF-αand RT-PCR to observe the expression of MBL1 mRNA. Results:Com-pared with the untreated diabetic group, urinary protein excretion, serum creatinine、serum urea nitrogen of GTW treated rats were substanitially decreased. The level of MBL-A、IL-6、TNF-α increased in DN group and then decreased when treated with GTW. Conclusion:GTW can reduce the injury of immune inflammatory response in DN rats, partly through down-regulating the expression of MBL-A、IL-6 and TNF-α.
