以脂肪酶为原料，通过单因素试验对影响脂肪酶固定化的因素进行分析，并运用响应面方法优化最佳工艺条件。结果表明：硅烷化试剂添加量0.35%、硅藻土与酶的质量比3.5∶1、温度29℃和时间4.5 h为脂肪酶的固定化最佳工艺组合，固定后的脂肪酶活力为2206.67 U·g-1。

Used lipase as raw material,the single factor experiment on affecting lipase immobilization was analyzed,and the response surface methodology was used to optimize the optimum conditions. The results showed that the amount of silylating agent 0.35%, mass ratio of diatomite and enzyme 3.5∶1,temperature 29℃and 4.5 h were combination of optimal lipase immobilization technology, and the activity of immobilized lipase was 2 206.67 U·g-1.
