目的:探讨链脲佐菌素(Streptozotocin ,STZ)诱导的糖尿病大鼠坐骨神经传导速度(NCV)变化,初步确立糖尿病周围神经病(diabetic peripheral neuropathy ,DPN)大鼠模型的构建和评价方法。方法:选用SD大鼠,随机分为正常对照组和模型组,模型组腹腔注射STZ。检测动物体质量、血糖、触觉、神经传导速度(NCV)变化,并取大鼠坐骨神经组织进行 HE染色,观察其形态结构变化。结果:与正常对照组比较,给予STZ后,模型组动物体质量逐渐下降,4 w 后比较差异有显著意义( P<0.05),血糖显著升高(P<0.001);50%机械缩足阈值,于注射STZ 2 w 后明显下降(P<0.05),注射STZ后4 w明显上升(P<0.05);给予STZ后4 w ,模型组大鼠运动神经传导速度(MCV)和感觉神经传导速度(SCV)显著减慢(P<0.05),坐骨神经组织神经纤维松散,密度不均匀。结论:注射STZ 4 W后,大鼠MCV和SCV减慢,触觉敏感度呈双相变化,坐骨神经出现病理形态结构,成功复制出DPN大鼠模型。

목적:탐토련뇨좌균소(Streptozotocin ,STZ)유도적당뇨병대서좌골신경전도속도(NCV)변화,초보학립당뇨병주위신경병(diabetic peripheral neuropathy ,DPN)대서모형적구건화평개방법。방법:선용SD대서,수궤분위정상대조조화모형조,모형조복강주사STZ。검측동물체질량、혈당、촉각、신경전도속도(NCV)변화,병취대서좌골신경조직진행 HE염색,관찰기형태결구변화。결과:여정상대조조비교,급여STZ후,모형조동물체질량축점하강,4 w 후비교차이유현저의의( P<0.05),혈당현저승고(P<0.001);50%궤계축족역치,우주사STZ 2 w 후명현하강(P<0.05),주사STZ후4 w명현상승(P<0.05);급여STZ후4 w ,모형조대서운동신경전도속도(MCV)화감각신경전도속도(SCV)현저감만(P<0.05),좌골신경조직신경섬유송산,밀도불균균。결론:주사STZ 4 W후,대서MCV화SCV감만,촉각민감도정쌍상변화,좌골신경출현병리형태결구,성공복제출DPN대서모형。
Objective:To investigate the changes siatic nerve conduction velocity in diabetic rats in‐duced by streptozotocin so as to establish and evaluate experimental diabetic peripheral neuropathy model rats .Methods:SD rats were randomly divided into 2 groups:the control group ,and the model group .Rats in the model group were injected intraperitoneally with STZ .Body weight ,fasting blood glucose and 50%of mechanical withdraw threshold were measured on time .Nerve conduction velocity was tested at the end of the experiment .Sciatic nerve was stained with H&E to observe the pathological changes .Results:Com‐pared with the control group ,rats in the DPN group showed significant decrease in weight (P<0 .05)3 weeks after injection of STZ and increase in fasting blood glucose (P<0 .05) 3 days after injecting STZ 50% mechanical withdraw threshold decreased significantly 2 weeks after injection of STZ (P< 0 .05) and decreased significantly 4 weeks after injection of STZ(P < 0 .05) .Motor nerve conduction velocity and sensory nerve conduction velocity declined significantly (P<0 .05) .Fibers of sciatic nerves arranged loosely ,in which the density was uneven .Conclusion:The experimental diabetic peripheral neuropathy model can be established by injecting with STZ ,tactile or pain sensitivity increases 2 weeks later and then decreases 4 weeks later ,and nerve conduction velocity and density of fibers of sciatic nerves decrease later 4 w eeks .
