目的:比较2种大肠杆菌培养方法的优劣。方法取大肠杆菌标准菌株[ ATCC25922]在2012年10-11月内分5次,每次20遍适当倍数的稀释,最终取得10-7、10-8、10-93个合适的稀释度共300分样液后,运用方法1取上述阳性对照样液上清液各1 ml接种单料乳糖胆盐发酵管3管进行检测;同时运用方法2测定剩余的上述样液,倒入双料乳糖胆盐发酵培养液中,置(36±1)℃培养24 h,对其进行平行比对实验。结果方法1阳性数为229件,方法2阳性数为249件,经χ2检验,2种大肠杆菌培养方法差异有统计学意义(P<0.05)。结论方法2比方法1简便、廉价、检出率高。建议将大肠菌群检验标准DB31/8-2004《托幼机构环境、空气、物体表面卫生要求及检测方法》中“按方法1:取样液上清液接种单料乳糖胆盐发酵管3管”修订为“按方法2:用测定剩余的检样,倒入双料乳糖胆盐发酵培养液中”。
目的:比較2種大腸桿菌培養方法的優劣。方法取大腸桿菌標準菌株[ ATCC25922]在2012年10-11月內分5次,每次20遍適噹倍數的稀釋,最終取得10-7、10-8、10-93箇閤適的稀釋度共300分樣液後,運用方法1取上述暘性對照樣液上清液各1 ml接種單料乳糖膽鹽髮酵管3管進行檢測;同時運用方法2測定剩餘的上述樣液,倒入雙料乳糖膽鹽髮酵培養液中,置(36±1)℃培養24 h,對其進行平行比對實驗。結果方法1暘性數為229件,方法2暘性數為249件,經χ2檢驗,2種大腸桿菌培養方法差異有統計學意義(P<0.05)。結論方法2比方法1簡便、廉價、檢齣率高。建議將大腸菌群檢驗標準DB31/8-2004《託幼機構環境、空氣、物體錶麵衛生要求及檢測方法》中“按方法1:取樣液上清液接種單料乳糖膽鹽髮酵管3管”脩訂為“按方法2:用測定剩餘的檢樣,倒入雙料乳糖膽鹽髮酵培養液中”。
목적:비교2충대장간균배양방법적우렬。방법취대장간균표준균주[ ATCC25922]재2012년10-11월내분5차,매차20편괄당배수적희석,최종취득10-7、10-8、10-93개합괄적희석도공300분양액후,운용방법1취상술양성대조양액상청액각1 ml접충단료유당담염발효관3관진행검측;동시운용방법2측정잉여적상술양액,도입쌍료유당담염발효배양액중,치(36±1)℃배양24 h,대기진행평행비대실험。결과방법1양성수위229건,방법2양성수위249건,경χ2검험,2충대장간균배양방법차이유통계학의의(P<0.05)。결론방법2비방법1간편、렴개、검출솔고。건의장대장균군검험표준DB31/8-2004《탁유궤구배경、공기、물체표면위생요구급검측방법》중“안방법1:취양액상청액접충단료유당담염발효관3관”수정위“안방법2:용측정잉여적검양,도입쌍료유당담염발효배양액중”。
Objective To compare the advantages and disadvantages of 2 Escherichia coli culture methods , so as to establish and improve disinfection and control in all the kindergartens of the area .Methods The standard strain of Escherichia coli [ ATCC25922] was collected in October and November of 2012, then, was diluted 10 times and each time with a proper ratio .Finally, 300 samples of solution were obtained with proper dilution levels of 10 -7 , 10 -8 and 10 -9 .Then, 1ml of the above positive control su-pernatant was inoculated in the unblended lactose bile salt fermentation tubes (3 tubes) by using method 1 for later detection .At the same time, detection was made in the remaining supernatant by using method 2, then, the test solution was put in the lactose bile salt fermentation culture solution , and was inoculated at a temperature of ( 36 ±1 )℃ for 24 hours for parallel comparison experiments . Results Positive detection rate of method 1 was 229, while that of method 2 was 249.Byχ2 test, statistical difference could be noted in the 2 Escherichia coli culture methods (P<0.05).Conclusion Method 2 was simpler and more economic and seemed to have a higher detection rate , when compared with that of method 1.It was recommended that test method 2 could replace method 1 in the Esch-erichia coli test standard DB31/8-2004.
