目的:观察miRNA Let‐7和Lin28在鼻咽癌组织中的相对表达并分析其相关性,探讨该调节环在鼻咽癌病理机制中的可能作用和意义。方法以正常鼻咽组织为对照,运用实时定量聚合酶链反应(Real‐time PCR)方法分别检查鼻咽癌组织中Lin28A/B和Let‐7a miRNA的相对表达量,并对两组数据进行相关性分析。采用免疫组织化学技术检查Lin28 A/B在鼻咽癌组织中的表达部位和水平。结果 Real‐time PCR实验显示,Lin28 miRNA在鼻咽癌组织中的相对表达量较对照组升高,而Let‐7a miRNA在鼻咽癌组织的相对表达量较对照组降低,差异有统计学意义;鼻咽癌组织中Lin28A/B miRNA和Let‐7a miRNA的相对表达量呈负相关。免疫组织化学分析证实,Lin28蛋白在鼻咽癌组织呈阳性表达,定位于细胞质中;而在正常鼻咽组织中,Lin28无表达。结论在鼻咽癌组织中Lin28表达上调而Let‐7a miRNA表达下调,且二者呈显著的负相关,提示Let‐7/Lin28双向负反馈调节异常可能参与了鼻咽癌的发生发展。

목적:관찰miRNA Let‐7화Lin28재비인암조직중적상대표체병분석기상관성,탐토해조절배재비인암병리궤제중적가능작용화의의。방법이정상비인조직위대조,운용실시정량취합매련반응(Real‐time PCR)방법분별검사비인암조직중Lin28A/B화Let‐7a miRNA적상대표체량,병대량조수거진행상관성분석。채용면역조직화학기술검사Lin28 A/B재비인암조직중적표체부위화수평。결과 Real‐time PCR실험현시,Lin28 miRNA재비인암조직중적상대표체량교대조조승고,이Let‐7a miRNA재비인암조직적상대표체량교대조조강저,차이유통계학의의;비인암조직중Lin28A/B miRNA화Let‐7a miRNA적상대표체량정부상관。면역조직화학분석증실,Lin28단백재비인암조직정양성표체,정위우세포질중;이재정상비인조직중,Lin28무표체。결론재비인암조직중Lin28표체상조이Let‐7a miRNA표체하조,차이자정현저적부상관,제시Let‐7/Lin28쌍향부반궤조절이상가능삼여료비인암적발생발전。
Objective To investigate the expressions of miRNA Lin28/Let‐7 ,a bidirectional negative feedback loop ,in na‐sopharyngeal carcinoma(NPC) ,so as to evaluate their role in the pathogenesis of NPC.Methods With normal nasopharyngeal tissues as control ,the expressions of miRNA Lin28A/B and miRNA Let‐7a in NPC were detected respectively by using real‐time PCR technique in NPC tissues and the correlation analysis was performed.Immunohistochemistry(IHC)was used to detect the expression levels and locations of Lin28A/B protein in the NPC tissues. Results Real‐time PCR showed that the expression of miRNA Lin28 was significantly up‐regulated in NPC tissues ,and miRNA Let‐7a was significantly down‐regulated in NPC tis‐sues as compared with the control tissues. There was statistically significant difference in the expressions of miRNA Lin28 and miRNA Let‐7a between the tumor and control tissues. IHC analysis demonstrated positive expression of Lin28 in NPC tissues , which was located in the cytoplasm ,and negative expression of Lin28 in normal nasopharyngeal tissues.Conclusion miRNA Lin28 is up‐regulated and miRNA Let‐7a down‐regulated in NPC ,and there is a significant negative association between them ,which indicates that abnormal bidirectional negative feedback of Lin28/Let‐7 may be involved in the molecular pathogenesis of NPC.
