目的::分析2型糖尿病(T2DM)合并颈动脉粥样硬化(CAS)患者外周血单个核细胞(PBMCs)中衔接蛋白凋亡相关斑点样蛋白(ASC)mRNA 的表达及作用。方法:根据华盛顿大学 CAS 斑块超声分级标准将107例T2DM 患者分为单纯 T2DM 组(n=26)、T2DM 轻度斑块组(n=32)、T2DM 中度斑块组(n=38)和 T2DM 重度斑块组(n=11)。另选非 T2DM 的 CAS 患者作为单纯 CAS 组(n =35),以及体检健康者作为正常对照组(n =35)。采用实时荧光定量聚合酶链反应(RT-FQ-PCR)技术检测各组 PBMCs 中 ASC mRNA 表达水平。统计分析各组差异及 ASC mRNA 水平与 CAS 斑块严重程度的关系。结果:各病例组 ASC mRNA 表达水平显著高于正常对照组(P <0.05),T2DM 合并 CAS 组 ASC mRNA 表达水平显著高于单纯 CAS 组(P <0.01);ASC mRNA 表达水平轻度斑块组>中度斑块组>重度斑块组,即 ASC mRNA 表达水平与 T2DM 患者 CAS 斑块程度呈明显负相关(r=-0.43,P <0.05)。结论:ASC mRNA 表达增加可能是 T2DM 合并 AS 的发病因素和 CAS 斑块活跃性的指征。

목적::분석2형당뇨병(T2DM)합병경동맥죽양경화(CAS)환자외주혈단개핵세포(PBMCs)중함접단백조망상관반점양단백(ASC)mRNA 적표체급작용。방법:근거화성돈대학 CAS 반괴초성분급표준장107례T2DM 환자분위단순 T2DM 조(n=26)、T2DM 경도반괴조(n=32)、T2DM 중도반괴조(n=38)화 T2DM 중도반괴조(n=11)。령선비 T2DM 적 CAS 환자작위단순 CAS 조(n =35),이급체검건강자작위정상대조조(n =35)。채용실시형광정량취합매련반응(RT-FQ-PCR)기술검측각조 PBMCs 중 ASC mRNA 표체수평。통계분석각조차이급 ASC mRNA 수평여 CAS 반괴엄중정도적관계。결과:각병례조 ASC mRNA 표체수평현저고우정상대조조(P <0.05),T2DM 합병 CAS 조 ASC mRNA 표체수평현저고우단순 CAS 조(P <0.01);ASC mRNA 표체수평경도반괴조>중도반괴조>중도반괴조,즉 ASC mRNA 표체수평여 T2DM 환자 CAS 반괴정도정명현부상관(r=-0.43,P <0.05)。결론:ASC mRNA 표체증가가능시 T2DM 합병 AS 적발병인소화 CAS 반괴활약성적지정。
Objective:To analysis expression and role of adaptor protein apoptosis-associated speck-like pro-tein containing a CARD (ASC)mRNA in peripheral blood mononuclear cells (PBMCs)of type 2 diabetes mellitus (T2DM)merger with carotid atherosclerosis (CAS)patient.Method:107 T2DM patients were enrolled,divided them into T2DM group (n = 26 ),T2DM small plaque group (n = 32 ),T2DM moderate plaque group (n = 38 ), T2DM serious plaque group (n = 1 1 )according to atherosclerosis plaque ultrasonic classification constituded by Washington university.Another non T2DM CAS patients as CAS group (n = 35 ),and healthy persons as normal control group (n=35).The ASC mRNA expression levels of PBMCs in each group was deteeted by real-time fluo-rescent quantitative polymerase chain reaction (RT-FQ-PCR)technology.Statistical analysis of the differences be-tween each group and relationship between ASC mRNA levels and the severity of CAS patches.Results:Expression level of ASC mRNA was significantly higher in the pathological group than normal control group (P <0.05 ),ex-pression level of ASC mRNA was significantly higher in T2DM combined with CAS group than CAS group (P <0. 01),ASC mRNA expression in small plaque group greater than moderate plaque group and severe plaque group,ex-pression of ASC mRNA was negatively correlated with CAS plaque degree in T2DM patients(r=-0.43,P <0.05). Conclusion:Increased expression of ASC mRNA may be risk factor of T2DM combined with AS and CAS.
