目的::探讨白细胞介素-10(IL-10)基因592A/C 位点多态性与结核病易感性的关系。方法:检索PubMed、Medline、EMbase 等数据库,搜集相关病例对照研究文献。文 献 经 筛 选 和 方法 学 质 量 评 价 后,采用 Sta-ta12.0软件进行 Meta 分析,计算合并 OR 值及其95%CI,最后进行敏感性分析及发表偏倚评估。结果:共16篇文献纳入研究,包括结核病患者(病例组)4115例,健康体检者(对照组)5441例。Meta 分析显示,在各个遗传模型中,IL-10-592A/C 基因多态性与结核病总体发病风险关系不大。对纳入研究以世界各洲为分层因素进行分析,在亚洲人群中等位基因 A 者结核病发病率高于等位基因 C 者(OR=1.26,95%CI=1.08-1.28,P <0.05);纯合子基因型 AA 者结核病发病率高于纯合子基因型 CC 者(OR=1.50,95%CI=1.07-2.12,P <0.05);纯合子基因型 AA 者结核病发病率高于 AC+CC 基因型者(OR=1.33,95%CI=1.10-1.62,P <0.05)。经 Begg’s 与 Egg-er’s 检验,纳入的所有研究未见明显发表偏倚。结论:IL-10-592A/C 基因多态性中等位基因 A 可能与亚洲人群结核病易感性风险增高有关。

Objective:To investigate the association between interleukin-10 (IL-10)gene promoter -592A/C polymorphism and tuberculosis(TB)susceptibility.Method:Such databases as PubMed,MEDLINE and EMbase data were searched to collect the case-control studies published.According to the inclusion and exclusion criteria, the studies were screened,the data were extracted,and the methodological quality of the included studies was evalu-ated.Then meta-analysis was conducted using Stata 12.0 sotware,the pooled odds ratio (ORs)with 95% conidence interval (CI)were calculated,and the sensitivity and publication bias were evaluated at the same time.Results:A to-tal of 1 6 studies were included,which involved 41 1 5 cases and 5441 healthy controls.The results of meta-analysis showed that,the IL-10 -592A/C polymorphism had no association with a increased risk of TB.In the stratified a-nalysis by ethnicity,significantly increased risk of TB was associated with Asians in IL-10 -592A/C polymorphism (A vs.C:OR=1.26,95% CI=1.08-1.28,P <0.05;AA vs.CC:OR=1.50,95% CI= 1.07-2.12,P <0.05;AA vs.AC+CC:OR=1.33,95% CI=1.10-1.62,P <0.05).All of the included studies showed no publication bias by Begg's and Egger's test.Conclusion:IL-10 -592A/C polymorphism in allele A might be a genetic risk factor that increases TB susceptibility for Asians.
