为建立一种同时测定海马中5种核苷(胞苷、尿苷、肌苷、鸟苷和腺苷)和5种核苷酸( CMP、 UMP、 IMP、 GMP和AMP)含量的方法,本研究采用HPLC,以Accalaim C30(2.1 mm ×150 mm,3μm)为分析柱,以水(A)-100 mmol/L NH4OAC溶液(pH=5.0)(B)-乙腈(C)为流动相,梯度洗脱,流速0.4 mL/min,检测波长254 nm,柱温25℃。结果显示,5种核苷和5种核苷酸的浓度均在0.02~10 mg/L范围内线性关系良好。平均加样回收率在88.38%~96.30%( n=3)之间。本法能够快速、简便地测定海马中主要核苷和核苷酸的含量,为其质量控制提供保证。

위건립일충동시측정해마중5충핵감(포감、뇨감、기감、조감화선감)화5충핵감산( CMP、 UMP、 IMP、 GMP화AMP)함량적방법,본연구채용HPLC,이Accalaim C30(2.1 mm ×150 mm,3μm)위분석주,이수(A)-100 mmol/L NH4OAC용액(pH=5.0)(B)-을정(C)위류동상,제도세탈,류속0.4 mL/min,검측파장254 nm,주온25℃。결과현시,5충핵감화5충핵감산적농도균재0.02~10 mg/L범위내선성관계량호。평균가양회수솔재88.38%~96.30%( n=3)지간。본법능구쾌속、간편지측정해마중주요핵감화핵감산적함량,위기질량공제제공보증。
In order to establish a method for the determination of nucleosides ( cytidine, uridine, inosine, guanosine and adenosine) and nucleotides ( CMP, UMP, IMP, GMP and AMP) in hippocampus, the simultaneous determination was performed on an Accalaim C30 (2.1 mmí150 mm, 3 μm) column by HPLC. Gradient elution was performed with water (A), 100 mmol/L NH4OAC solution (adjusted pH to 5.0)(B)-acetonitrile(C) as the mobile phase. The flow rate was 0.4 mL/min, the system was performed at 25 ℃ and the nucleosides and nucleotides were monitored and quantified at a wavelength of 254 nm. The five nucleosides and the five nucleotides showed good linearity with their concentrations in the range of 0.02~10 mg/L. Their average recoveries were between 88.38% and 96.30%. The HPLC method established in this study is a rapid and simple method for the simultaneous determination of nucleosides and nucleotides in Hippocampus and is helpful to its quality control.
