目的:探讨肿瘤坏死因子相关凋亡诱导配体(TRAIL)联合三氧化二砷(ATO)对裸鼠肺癌移植瘤的抑制作用及其可能机制。方法将裸鼠肺癌移植瘤模型随机分为阴性对照组、TRAIL组、ATO低剂量组(ATO1组)、ATO高剂量组(ATO2组)、TRAIL联合ATO组,称取瘤质量并计算抑瘤率,采用免疫组化法检测半胱氨酸天冬氨酸蛋白酶3(Caspase-3)的表达。结果与阴性对照组比较,ATO1组、ATO2组对裸鼠肺癌移植瘤均具有抑制作用,TRAIL联合ATO组具有协同增效作用,并且上调Caspase-3的表达,差异均有统计学意义(P<0.05)。结论 ATO能增强TRAIL对裸鼠肺癌移植瘤的抑制作用,其机制可能与促进Caspase-3表达有关。

목적:탐토종류배사인자상관조망유도배체(TRAIL)연합삼양화이신(ATO)대라서폐암이식류적억제작용급기가능궤제。방법장라서폐암이식류모형수궤분위음성대조조、TRAIL조、ATO저제량조(ATO1조)、ATO고제량조(ATO2조)、TRAIL연합ATO조,칭취류질량병계산억류솔,채용면역조화법검측반광안산천동안산단백매3(Caspase-3)적표체。결과여음성대조조비교,ATO1조、ATO2조대라서폐암이식류균구유억제작용,TRAIL연합ATO조구유협동증효작용,병차상조Caspase-3적표체,차이균유통계학의의(P<0.05)。결론 ATO능증강TRAIL대라서폐암이식류적억제작용,기궤제가능여촉진Caspase-3표체유관。
Objective To approach the inhibitory action and proposed mechanism of TRAIL combined with ATO on transplanted human lung cancer in nude mice. Methods The nude mice bearing A549 lung cancer samples were randomly divid-ed into the negative control group,group TRAIL,group ATO1,group ATO2 and TRAIL+ATO group. Tumor tissues were weighted and the inhibitory rate was observed. Immunohistochemistry was applied to detect the expression of Caspase-3 after treatments. Re-sults Compared with the negative control group,it had a strong the obviously action inhibition on transplanted human lung cancer of nude mice in group ATO1 and group ATO2 while a synergistic effect in group TRAIL+ATO ,and increased the expression of Caspase-3,whose difference had statistical significance(P<0.05). Conclusion ATO may inhibit the growth of transplanted hu-man lung cancer in nude mice through strengthening TRAIL,whose possible mechanism is potentially related to the expression of Caspase-3.
