目的：在新疆特高发肿瘤资源库中建立冷冻组织质量鉴定体系。方法选取肿瘤资源库中食管癌、宫颈癌、肝癌冷冻组织及正常对照组织各10例，利用 HE 染色技术检测标本取材的准确性，琼脂糖凝胶电泳鉴定DNA 质量，琼脂糖凝胶电泳检测 RNA 质量，Western-blot 鉴定蛋白质质量。结果 HE 染色观察检测标本肿瘤部分占80％以上，28s／18s 为1．8～2．0，蛋白质及 DNA 质量均完整无降解。结论建立了一个规范化的冷冻组织质量控制体系，可以作为肿瘤资源库标本质量鉴定的常规方法。
목적：재신강특고발종류자원고중건립냉동조직질량감정체계。방법선취종류자원고중식관암、궁경암、간암냉동조직급정상대조조직각10례，이용 HE 염색기술검측표본취재적준학성，경지당응효전영감정DNA 질량，경지당응효전영검측 RNA 질량，Western-blot 감정단백질질량。결과 HE 염색관찰검측표본종류부분점80％이상，28s／18s 위1．8～2．0，단백질급 DNA 질량균완정무강해。결론건립료일개규범화적냉동조직질량공제체계，가이작위종류자원고표본질량감정적상규방법。
Objective To establish a standard protocol for quality control of frozen tissues in the Xinjiang-Tumor Resource Bank.Methods Ten frozen tissues and normal control tissues of cervical cancer esophagus cancer and hepatocellular carcinoma were respectively randomly selected,stained with HE and pathologically checked.Agarose electrophoresis was used to detect the quality of the DNA.Agarose elec-trophoresis used to check the RNA quality.Western blot was used to detect the quality of the protein from these frozen tissues.Results HE staining showed that all the detected samples contained 80% or greater tumor.28s/18s ratio were respectively 1.8-2.0.The good quality of the protein and DNA from these fro-zen tissues was observed.Conclusion We have established a standard quality control system,which is ap-plicable as a routine method in quality control of frozen tissues from the tumor resource bank.
