为利用 ISSR 分子标记进行剑麻遗传多样性分析和种质资源鉴定，分析了剑麻 ISSR 反应的主要参数对反应体系的影响。结果表明：25μL 的反应体系中含150 ng 的模板 DNA、10μLTaq 聚合酶MIX、0．5μmol／L 引物。优化的反应程序：94℃预变性5 min；94℃变性30 s，设定温度（51～60．3℃）退火45 s，72℃延伸45 s，循环35次；然后72℃延伸10 min；筛选到16条适合剑麻扩增的 ISSR 引物。

The effects of main parameters on ISSR reaction system in sisal was studied to conduct genetic diversity analysis and germplasm resources identification by using ISSR molecular marker.Results:In a total volume of 25 L ISSR system,it contains 150 ng temple DNA,0.5 μmol/L primer,10 μL Taq DNA polymerase MIX;The suitable PCR procedure is one cycle denaturing at 94℃ for 5 min;35 cycles each involved denaturing at 94℃ for 30s,annealing at the set temperature (51 ~ 60.3℃)for 45 s, extending at 72℃ for 45 s,one cycle extending at 72℃ for 10 min,then remaining at 4℃.Based on this optimized system,16 primers were screened.
