为制备与民间治疗带状疱疹用烟草烟油疗效相似的烟草提取物,采用高效液相色谱法对民间不同来源的烟草烟油进行指纹图谱构建,并筛选其制备工艺。结果表明:在色谱柱为 Inertsil ODS-SP (4.6×250 mm,5μm),流动相为甲醇-磷酸缓冲液(10∶90),检测波长为259 nm,柱温为35℃的条件下,测得5批不同来源烟草烟油有4个共有峰,方法学考察及相似度评价符合指纹图谱技术要求;利用该指纹图谱对冷浸、热回流、干馏、超声等4种工艺制备的烟草提取物进行评价,特征峰面积比较显示,超声法为制备烟草提取物适宜的方法。

In order to prepare the tobacco extract with similar efficacy to folk tobacco oil in treatment of herpes zoster,the method of HPLC was applied to establish a chromatographic fingerprint of tobacco oil from different origins in folk and to screen the preparation process of tobacco extract.The results showed that the five batches of tobacco oil had four common peaks under the condition that the separation was performed on a Inertsil ODS-SP column (4.6×250 mm,5 μm)with a mobile phase consisting of methanol and phosphate buffer solution (10∶90)and the detection was carried out on a UV detector at 259 nm and a column temperature at 35℃,the results of methodological study and similarity evaluation met the requirement of chromatographic fingerprint.According to the fingerprint of folk tobacco oil,the tobacco extracts prepared through four different preparation processes (cold soak,hot reflux,distillation and ultrasound)were evaluated.The results indicated that the ultrasound method was more suitable than other methods for preparation of tobacco extract by comparison of the characteristic peaks area.
