为去除烟梗细胞壁物质,改善烟梗抽吸品质,选取纤维素酶和果胶酶,采用正交试验优化超声波耦合酶反应去除烟梗中纤维素和果胶的工艺。结果表明:各因素对烟梗果胶降解率、纤维素降解率有显著影响(P <0.05),超声时间对去除烟梗中果胶、纤维素含量及感官质量的影响均较大,其贡献率分别达72.80%、32.59%、40.55%。经优化分析,超声波辅助复合酶去除烟梗中纤维素和果胶的较佳工艺条件为纤维素酶浓度50 U/L、果胶酶浓度600 U/L、超声功率400 W、超声时间50 min。优化后烟梗中果胶和纤维素的去除率分别达69.3%和31.5%,还原糖含量显著升高(P <0.05),总植物碱和总氮含量显著降低(P <0.05),感官质量明显改善,表皮结构更加疏松。

To remove cell wall materials of tobacco stems and improve sensory quality,cellulose and pectin were digested by cellulase and pectinase through L9 (3 4 )orthogonal optimization design under ultrasonic assisted.The results showed that each factor distinctly affect (P <0.05)digestion rate of pectin and cellulose,and the time of ultrasound is the most important factor,its contribution rate to digestion of pectin and cellulose,and improvement of sensory quality are 72.80%,32.59% and 40.55%,respectively. Through the optimization analysis,the optimal conditions to digestion of pectin and cellulose in tobacco stem are as follows:cellulase 50 U/L,pectinase 600 U/L,ultrasonic power 400 W and total extracting time 50 minutes.Some chemical components content in tobacco stems were changed after treated under the optimal conditions,pectin and cellulose reduced by 69.3% and 31.5%,respectively,reducing sugar rose observably (P <0.05),total alkaloid and total nitrogen decreased significantly(P <0.05),and the sensory quality markedly improved,skin structure changed to loose.
