为了探讨黄芪水煎液能否诱导大鼠肠黏膜微血管内皮细胞(RIMVECs)表达IFN-γ,采用MTT和双夹心ELISA分别检测了黄芪水煎液对体外培养的 RIMMVECs增殖以及表达 IFN-γ的影响。结果显示,黄芪水煎液在25 mg/L~3200 mg/L时促进 RIMVECs增殖。为更好的研究黄芪水煎液诱导效果,将不同浓度的黄芪水煎液(0.1、1、6、30 mg/mL)诱导 RIMVECs 6、12、24 h检测 IFN-γ含量,结果显示,在各检测时间点0.1、1 mg/mL组和空白组差异显著(P<0.05),6 mg/mL组差异极显著(P<0.01),30 mg/mL组差异不显著(P>0.05)。说明黄芪水煎液的抗病毒作用可能与诱导微血管内皮细胞表达 IFN-γ有直接的关系,为黄芪“益气解毒”功效的现代阐释提供了试验依据。

To study the effect of Astragalus aqua on interferon-γ(IFN-γ)expression in intestinal microvascular endo-thelial cells (IMMVECs),rat intestinal mucosa microvascular endothelial cells (RIMMVECs)were co-cultured with Astragalus aqua,and the cell proliferation and IFN-γexpression level were examined by MTT and sandwich ELISA, respectively.The results showed that the proliferation of RIMMVECs was increased by Astragalus aqua at different concentrations ranged between 25 mg/L and 3 200 mg/L.The effect of IFN-γexpression induced by Astragalus aqua was further explored;RIMMVECs were co-cultured with Astragalus aqua at concentrations of 0.1,1,6,30 mg/mL for 6,12,24 hours,respectively.The expression of IFN-γwas significantly increased by Astragalus aqua at concentrations of 0.1,1 mg/mL at each time point compared to that of blank group.And the expression of IFN-γwas even more sig-nificantly increased by Astragalus aqua at concentration of 6 mg/mL;whereas,no significant increase by Astragalus aqua was observed at concentration of 30 mg/mL compared to that of blank control.Our findings indicated that the anti-virus effect of Astragalus aqua might be due to its induction effect on microvascular endothelial cells to produce IFN-γ,which offered the molecular biology evidence to interpret its'immune-resistance enhancing and disintoxication'effect as an ancient Chinese herb.
