目的:利用效应面法优选白芷的醇提取工艺参数。方法:采用HPLC方法,Waters公司的 Sun-fire C18色谱柱(150 mm×4.6mm,5μm);流动相为甲醇-水(70∶30),等度洗脱;流速0.9 mL/min;检测波长254 nm;柱温30℃;分析时间为20分钟;进样量10μL。结果:白芷中欧前胡素的最佳提取工艺为甲醇浓度83.17%,提取时间9.15小时,甲醇用量为147.32 mL;异欧前胡素的最佳提取工艺为甲醇浓度91.83%,提取时间9.15小时,甲醇用量为112.68 mL。结论:采用星点设计与效应面法相结合的方法对实验工艺优化更方便、更准确。

Objective:To collect the extracting parameters of the alcohol from Baizhi (angelica dahurica) by central composite design (CCD) and response surface method (RSM). Methods:The HPLC method was performed and the detected conditions were defined, chromatography column:Waters' Sunfire C18 (150 mm×4.6mm, 5μm);mobile phase:CHOH/H2O=70/30, isocratic elution;flow rate:0.9 mL/min;detection wavelength:254 nm;column temperature:30℃;analyzing time:20 min;injection volume:10μL. Results:The best concentration of methanol for extracting the imperatorin from Baizhi was 83.17%, the extracting time was 9.15 hours and the methanol dosage was 147.32 mL,while for extracting the isoimperatorin the best concentration of methanol was 91.83%with 9.15 extract-ing hours and 112.68 mL methanol dosages. Conclusion: The method of combining CCD and RSM is easier and more accurate for optimizing the extracting process.
