DanHong injection%testicular torsion%SOD%NOS%MDA
目的:探讨丹红注射液对大鼠单侧睾丸扭转复位后健侧睾丸的组织学以及附睾精子生成的影响。方法:将30只健康雄性Wistar大鼠随机分为假手术组(A组)、睾丸扭转复位组(B组)、丹红注射液处理组(C组),每组10只。按Turner法建立左侧睾丸扭转模型,A组大鼠切开左侧阴囊,使左侧睾丸充分游离暴露,不予以扭转,模拟Turner法手术过程随意搬动睾丸,随后缝合关闭阴囊;B组大鼠按Turner法制作睾丸扭转模型:左侧睾丸顺时针绕精索旋转720°,肉膜白膜缝合固定以防止复位,阴囊缝合,扭转6小时后复位固定;C组大鼠按上述相同方法制备睾丸扭转模型,于睾丸扭转复位前60分钟腹腔注射丹红注射液(2 mL/kg),同时在术后3天每天分别注射丹红注射液(2 mL/kg)。各组喂养至术后4周时处死,取右侧睾丸标本行组织病理学观察,同时取右侧附睾尾部行精子计数及活动率检测。检测睾丸组织一氧化氮合酶(NOS)、血清超氧化物歧化酶(SOD)和丙二醛(MDA)水平。结果:C组与B组相比,生精上皮层数增多,平均曲细精管直径(MSTD)增大,同时附睾精子计数与活动率明显升高,其差异有统计学意义(P<0.05)。 C组大鼠睾丸组织SOD水平升高,NOS、MDA水平降低,差异有统计学意义(P<0.05)。结论:丹红注射液能够减轻大鼠单侧睾丸扭转复位后健侧睾丸组织损害,使附睾精子生成显著增多,机制可能为减少大鼠睾丸组织内自由基生成,提高其抗自由基能力。

목적:탐토단홍주사액대대서단측고환뉴전복위후건측고환적조직학이급부고정자생성적영향。방법:장30지건강웅성Wistar대서수궤분위가수술조(A조)、고환뉴전복위조(B조)、단홍주사액처리조(C조),매조10지。안Turner법건립좌측고환뉴전모형,A조대서절개좌측음낭,사좌측고환충분유리폭로,불여이뉴전,모의Turner법수술과정수의반동고환,수후봉합관폐음낭;B조대서안Turner법제작고환뉴전모형:좌측고환순시침요정색선전720°,육막백막봉합고정이방지복위,음낭봉합,뉴전6소시후복위고정;C조대서안상술상동방법제비고환뉴전모형,우고환뉴전복위전60분종복강주사단홍주사액(2 mL/kg),동시재술후3천매천분별주사단홍주사액(2 mL/kg)。각조위양지술후4주시처사,취우측고환표본행조직병이학관찰,동시취우측부고미부행정자계수급활동솔검측。검측고환조직일양화담합매(NOS)、혈청초양화물기화매(SOD)화병이철(MDA)수평。결과:C조여B조상비,생정상피층수증다,평균곡세정관직경(MSTD)증대,동시부고정자계수여활동솔명현승고,기차이유통계학의의(P<0.05)。 C조대서고환조직SOD수평승고,NOS、MDA수평강저,차이유통계학의의(P<0.05)。결론:단홍주사액능구감경대서단측고환뉴전복위후건측고환조직손해,사부고정자생성현저증다,궤제가능위감소대서고환조직내자유기생성,제고기항자유기능력。
Objective:To study the effect of DanHong injection on the histology of healthy testis and the sperm production of epididymides of rats undergoing the reduction by testicular torsion on the affected side. Methods:The 30 male Wistar rats were randomized into the sham-operated group (A), the reduction group (B) and the injection group (C) with 10 rats in each. The models with testicular torsion established by Turner method, rats in group A were cut the left scrotum to isolate and expose the testis without torsion and rotate the testis by imitating the Turner operating process and lastly stitched the wound; as in the a group the models established, the left testis of rats in group B were rotated 720° clockwise round the varicosity to be fixed by stitching the sarcolemma and albuginea and the left scrotum were closed, after fixation for 6 hours the testis were reduced and fixed;the models established as in the A and B group, the testis in the C group were injected DanHong injection 2 mL/kg in the abdomen before the re-duction of torsion for 60 minutes and after the operation the rats were injected DanHong injection 2 mL/kg for 3 days. After feeding for 4 weeks, the rats of three groups were killed, the right testis were collected to be observed histopathologically, the sperm of the right epididymides were counted and their activity ratio were detected. Mean-while, the levels of the nitricoxide synthase (NOS), serum superoxide dismutases (SOD) and malondialdehyde (MDA) were determined respectively. Results:In the comparison of the C and B group, the quantities of seminifer-ous epithelium cell of the rats in group C were increased, the diameters of the seminiferous tubules were enlarged, sperm counts of epididymides and activity ratios were raised significantly, and the differences between the two groups were significant (P<0.05). The SOD levels of the rats' testes in group C were increased, while the NOS and MDA levels were decreased, and the differences showed statistical meaning (P<0.05). Conclusion:The DanHong injection can lighten impairment of the healthy testis from the reduction of torsion of the affected side and signifi-cantly increase the sperm production of epididymides, one of the mechanisms may be relevant to its reducing the production of the free radicals and advancing its ability in fighting against the free radicals.
