目的:探讨巴马小型猪2型糖尿病( T2DM)模型胰岛素抵抗( IR)及瘦素、抵抗素水平的变化。方法选用健康雄性巴马小型猪10只,随机分为两组各5只。对照组喂养普通饲料;模型组喂养高脂高糖饲料,共喂养10个月。11个月初模型组腹腔注射链脲佐菌素(STZ)100 mg/kg,1周后重复1次;对照组腹腔注射等剂量柠檬酸—柠檬酸钠缓冲液,12个月末试验结束。检测两组空腹血糖( FPG)、空腹胰岛素( FINS)及血清瘦素、抵抗素水平,同时采用HOMA模型公式计算胰岛素抵抗指数( HOMA-IR)。结果与对照组比较,高脂高糖饲料喂养10个月模型组小型猪体质量、FPG、FINS及HOMA-IR升高(P均<0.01);与对照组相比,模型组STZ给药后(12个月末试验结束时)FPG进一步升高,FINS水平明显下降(P均<0.01);与对照组比较,模型组血清瘦素、抵抗素水平升高(P均<0.05),且HOMA-IR与瘦素、抵抗素呈正相关(r分别为0.893、0.933, P均<0.05)。结论 T2DM小型猪血清瘦素、抵抗素水平升高,与HOMA-IR密切相关。

목적:탐토파마소형저2형당뇨병( T2DM)모형이도소저항( IR)급수소、저항소수평적변화。방법선용건강웅성파마소형저10지,수궤분위량조각5지。대조조위양보통사료;모형조위양고지고당사료,공위양10개월。11개월초모형조복강주사련뇨좌균소(STZ)100 mg/kg,1주후중복1차;대조조복강주사등제량저몽산—저몽산납완충액,12개월말시험결속。검측량조공복혈당( FPG)、공복이도소( FINS)급혈청수소、저항소수평,동시채용HOMA모형공식계산이도소저항지수( HOMA-IR)。결과여대조조비교,고지고당사료위양10개월모형조소형저체질량、FPG、FINS급HOMA-IR승고(P균<0.01);여대조조상비,모형조STZ급약후(12개월말시험결속시)FPG진일보승고,FINS수평명현하강(P균<0.01);여대조조비교,모형조혈청수소、저항소수평승고(P균<0.05),차HOMA-IR여수소、저항소정정상관(r분별위0.893、0.933, P균<0.05)。결론 T2DM소형저혈청수소、저항소수평승고,여HOMA-IR밀절상관。
Objective To establish Bama miniature pig with type 2 diabetes mellitus ( T2DM) model which induced by high fat and high sugar diet and intraperitoneal injection of streptozotocin (STZ), and to investigate the changes of insu-lin resistance and adipocytokines in the Bama miniature pig with type 2 diabetes mellitus model .Methods Firstly, 10 healthy male Bama miniature pigs were equally randomly divided into control group and diabetic group ( model group ) . Secondly , the pigs of control group was fed with normal diet , while the pigs of model group was fed with high fat and high carbohydrate.After feeding for 10 months, the model group was injected with STZ 100 mg/kg, the injection was repeated a week later, while the control group was injected with the same amount of sodium citrate solution .Finally, the observation of the laboratory targets including FPG , FINS, Leptin and Resistin were carried on .Meanwhile, the insulin resistance index ( HOMA-IR) was calculated .Results After the early stage of experiment , the levesl of weight , FPG, FINS, HOMA-IR increased significantly in the model group comparing with the control group (all P<0.01).After administration of STZ, the serum level of FPG increased significantly , and the level of FINS was lower in model group as compared with control group (all P<0.01).The levels of Leptin and Resistin were higher in the model group comparing with the control group (all P<0.05) and the correlation analysis showed that HOMA-IR was positively correlated with the levels of Leptin and Resistin in the model group (all P<0.05).Conclusion The levels of leptin and resistin are associated with the insulin resistance of T2DM miniature pig.
