Teriparatide%MGP%Ovariectomized rat%Vascular calcification
目的:观察特立帕肽对老年去卵巢大鼠血管钙化的影响,并探讨其发挥作用的分子机制。方法用30只10个月龄SD大鼠,随机分为3组院假手术组、去卵巢组及特立帕肽治疗组。特立帕肽治疗组给予20μg/kg特立帕肽皮下注射,隔天1次,持续12周。术前及给药期间收集大鼠血清,给药结束后观察子宫的组织学变化。酶联免疫吸附法(ELISA)检测血中未羧化基质羧基谷氨酸蛋白(uc-MGP)的量及雌激素的变化;原子分光光度计测量血管总钙含量;Von Kossa染色观察动脉钙化;免疫组化法观察血管uc-MGP的表达;荧光实时定量PCR扩增MGP mRNA,观察其在胸主动脉的表达水平。结果血清中uc-MGP含量去卵巢组最高,特立帕肽治疗组其次,假手术组最低(P<0.05)。去卵巢组及特立帕肽治疗组雌激素水平明显下降(P<0.05)。去卵巢组血管总钙含量明显高于假手术组(P<0.01),特立帕肽治疗组明显低于去卵巢组(P<0.01)。血管未羧化MGP表达出现在血管钙化周围区域,钙化严重组表达较多。特立帕肽治疗组阳性较少表达。去卵巢组表达量最低,假手术组MGP mRNA表达量最高,特立帕肽治疗组表达量介于两者之间,差异有统计学意义(P<0.01)。结论特立帕肽对绝经后血管钙化有改善作用,可能是通过调节MGP的基因表达及活性发挥作用。

목적:관찰특립파태대노년거란소대서혈관개화적영향,병탐토기발휘작용적분자궤제。방법용30지10개월령SD대서,수궤분위3조원가수술조、거란소조급특립파태치료조。특립파태치료조급여20μg/kg특립파태피하주사,격천1차,지속12주。술전급급약기간수집대서혈청,급약결속후관찰자궁적조직학변화。매련면역흡부법(ELISA)검측혈중미최화기질최기곡안산단백(uc-MGP)적량급자격소적변화;원자분광광도계측량혈관총개함량;Von Kossa염색관찰동맥개화;면역조화법관찰혈관uc-MGP적표체;형광실시정량PCR확증MGP mRNA,관찰기재흉주동맥적표체수평。결과혈청중uc-MGP함량거란소조최고,특립파태치료조기차,가수술조최저(P<0.05)。거란소조급특립파태치료조자격소수평명현하강(P<0.05)。거란소조혈관총개함량명현고우가수술조(P<0.01),특립파태치료조명현저우거란소조(P<0.01)。혈관미최화MGP표체출현재혈관개화주위구역,개화엄중조표체교다。특립파태치료조양성교소표체。거란소조표체량최저,가수술조MGP mRNA표체량최고,특립파태치료조표체량개우량자지간,차이유통계학의의(P<0.01)。결론특립파태대절경후혈관개화유개선작용,가능시통과조절MGP적기인표체급활성발휘작용。
Objective To explore the effect and mechanism of teriparatide on the vascular calcification in aged ovariec-tomized rats. Methods Thirty ten months female SD rats were allocated into three groups randoml:sham operation group,ovariectomized group and teriparatide treatment group.Subcutaneous injection with teriparatide (20 μg/kg,every other day)after two weeks of ovariectomy for 12 weeks was used in teriparatide treatment group.All rats were kept serum before ovariectomy and during the administration period.After administration,HE stain of the uterus.The content of MGP in serum was determined by enzyme-linked immunosorbent assay(ELISA).The content of arterial calcium was measured. Aterial calcification was evaluated by Von Kossa staining.Expression of uc-MGP was detected by immunochistochem-istry.Relative quantification of MGP mRNA expression in arterial was detected by fluorescent real-time quantitative re-verse-transcription polymerase chain reaction. Results The content of serum uc-MGP in ovariectomized group was higher than that in teriparatide treatment group and sham group respectively (P<0.05).The estrogen level in ovariec-tomized group and teriparatide treatment group was significantly decreased respectively (P<0.05).The content of arterial calcium in ovariectomized group was higher than that in sham group (P<0.01),the content of arterial calcium in teri-paratide treatment group was lower than that in ovariectomized group (P<0.01).Expression of vascular uncarboxylated MGP appeared in the area around the vascular calcification,calcification group expressed more serious.Teriparatide treatment group with less positive expression.The lowest expression of MGP mRNA in ovariectomized group,the expres-sion of MGP mRNA was highest in sham operation group,expression level of teriparatide treatment group between the two groups,with statistical difference (P<0.01). Conclusion Teriparatide can improve vascular calcification of post-menopause women possibly through the regulation of MGP carboxylation and gene expression.
